Outline of Research at the Start

加齢は発がんの危険因子であり、超高齢化社会を迎えた日本においても、H.pylori感染率低下や除菌療法の普及にも関わらず、依然、胃癌の罹患率は加齢とともに高くなっている。このため、高齢者発症の胃癌の分子生物学的特徴を理解することは臨床的に重要である。そこで、近年確立した、幹細胞を維持増殖させ臓器に近い分化を誘導できるオルガノイド細胞培養系を用い、ヒト胃癌より胃オルガノイドを樹立して、その発症機序を分子生物学的に解明する。

Outline of Final Research Achievements

Aging is considered one of the risk factors for gastric carcinogenesis. To clarify its mechanism with aging, a novel 3D cell culture system, “Organoid”, was employed in this study. We established gastric organoids from human differentiated intramucosal gastric cancer and created gene expression profiles. In gastric cancer-derived organoids, there was an activation of the Wnt/beta-catenin signaling pathway, induction of transcription factors that suppress cellular senescence, and an enhancement of cell proliferative capacity. These gene expression profiles largely matched those of gastric organoids derived from wild-type aged model mice, shedding light on part of the human gastric carcinogenesis with aging.

Academic Significance and Societal Importance of the Research Achievements

本研究で胃発癌過程の極初期の段階と考えられるヒト分化型粘膜内胃癌から胃オルガノイドを樹立し、正常の加齢マウスモデル由来胃オルガノイドの遺伝子発現プロファイルと比較することによって、加齢に伴い出現する細胞増殖能が高い病的幹細胞が発癌に関わることが類推できた。病態解明の一助となると同時に、超高齢社会の日本のおいて高齢発症の胃癌の悪性度や発癌リスクの予測に今後貢献することが期待された。
