Outline of Research at the Start

Toll-like receptor 9シグナルとブラジキニンB2受容体経路に着目した過敏性腸症候群の発症メカニズムを腸管知覚の評価や生物学的手法によって検討し、複雑な病態の解明を試みるとともに、新規治療法の開発を視野に入れ検証していく。

Outline of Final Research Achievements

A follow-up study after a waterborne gastroenteritis outbreak suggested involvement of specific genetic variants, including toll-like receptor (TLR)9. Based on these results, we investigated the relationship between TLR9 and PI-IBS development using an animal model. Mice lacking TLR9 showed significant intestinal hypersensitivity even after the infectious gastroenteritis improved, and it was found that the cause of this was increased expression of the bradykinin receptor, which is involved in pain perception.

Academic Significance and Societal Importance of the Research Achievements

本研究により、TLR9シグナル欠損状態に腸管感染が加わることで過敏性腸症候群を発症すること、その病態にブラジキニン受容体の発現亢進が関与していることが判明した。
また、その後の実験によりブラジキニン受容体の選択的拮抗薬を用いることで腸管知覚過敏が抑制されることが実証された。
感染後過敏性腸症候群は、いまだ病態に不明な点が多く、効果的な治療が乏しいのが現状であり、本研究は病態解明や治療法の開発へのBreak throughとなりうる。
