Outline of Research at the Start

申請者らは、細胞増殖・成長のmaster regulatorである哺乳類ラパマイシン標的蛋白質複合体1 (mTORC1) の恒常的活性化が、MLKL活性化依存性プログラムネクローシスであるネクロプトーシスを誘導することを報告した。さらに、リン酸化により活性化したMLKLが拡張型心筋症の心筋細胞で増加し、特に核リン酸化MLKL増加が不良な転帰と関係していた。本研究の目的は、心不全の発症・進展におけるMLKLの役割を解明し、MLKL標的治療の基盤を作ることである。

Outline of Annual Research Achievements

本年度はミトコンドリア機能障害とネクロプトーシスの関連について検証した。

1. H9c2ラット心筋芽細胞を用いてTNF-alphaとcaspase阻害薬同時投与(TNF/zVAD)により誘導されたネクロプトーシスがミトコンドリア形態に与える影響を検討した。TNF/zVAD投与4時間後にミトコンドリアが一時的に断片化したが、その後に融合し、12時間後には投与前と比較して有意に融合ミトコンドリアが増加した。ミトコンドリア融合の調節因子であるMfn1の蛋白量はvehicle群と比較してTNF/zVAD添加4時間後および12時間後に、Mfn2の蛋白量はTNF/zVAD添加1時間後に有意に増加していた。
2. TNF/zVAD添加18時間後の培養液中のLDH活性を指標としたネクロプトーシスの程度は、control siRNA導入群とDrp1 siRNA導入群あるいはFis1 siRNA導入群の間で同様であった。一方、TNF/zVAD添加18時間後の培養液中のLDH活性は、control siRNA導入群と比較してMfn1/2 siRNA導入群もしくはOpa1 siRNA導入群で有意に低下していた。以上から、ミトコンドリア融合蛋白発現抑制はネクロプトーシスを抑制することが明らかとなった。
3. control siRNA導入群では、TNF/zVAD添加4あるいは8時間後にRIP1-Ser166のリン酸化が増加していた。siRNA導入によるMfn1/2あるいはOpa1発現抑制によりTNF/zVADによるRIP1-Ser166のリン酸化が軽減した。以上から、ミトコンドリア融合蛋白発現低下によるRIP1-Ser166リン酸化軽減を介したRIP1活性化抑制がネクロプトーシス抑制の機序であることが明らかとなった。

2023年度に本研究の成果を論文として発表した。また、MLKLノックアウトマウスを用いた解析も現在進行中である。

Strategy for Future Research Activity

MLKLノックアウトマウス(MLKL-KO)とコントロール(WT)を用いてドキソルビシン心筋症モデルを作成した。MRIで評価した左室収縮機能はMLKL-KOでWTと比較して有意に改善していた。MLKL活性化がドキソルビシン心筋症の分子機構として重要な役割を果たす可能性があり、様々な分子生物学的手法を用いて解析している。
