申請者は酸化ストレス応答転写因子Nrf2 (NF-E2-related factor 2)の薬理学的活性化がマウスにおいて肥満を抑制することを報告した(Diabetes 2017)。さらに、そのメカニズムを探求する中で、Nrf2の薬理学的活性化が腸管からの脂肪吸収を制御する可能性を見出した(未発表)。本研究の目的は、脂肪の吸収を担う腸管上皮細胞に着目して、Nrf2と脂肪吸収をつなぐ分子メカニズムを解明し、Nrf2の活性化による脂肪吸収の制御が肥満の抑制にも寄与するかを明らかにすることである。本研究の成果は、Nrf2活性化薬を肥満症の治療に応用するための研究基盤となることが期待される。

| Outline of Final Research Achievements |
Previously, the applicant reported that glucoraphanin (GR), an activator of the transcription factor Nrf2 (Nuclear factor erythroid 2-related factor 2), acts on brown adipose tissue to increase energy expenditure through thermogenesis and suppress obesity in mice fed a high-fat diet (Nagata N et al., Diabetes, 2017). I further investigated how pharmacological activation of Nrf2 influences intestinal fat absorption. They found that both GR and bardoxolone methyl (BARD), another Nrf2 activator, showed anti-obesity effects by reducing body weight in mice made obese by a high-fat diet. Furthermore, an oral oil loading test revealed that both drugs suppressed fat absorption. This suggests that activating Nrf2 may play a role in inhibiting the absorption of fat from the intestines.

| Academic Significance and Societal Importance of the Research Achievements |
肥満の是正は、高血圧、脂質異常症、脂肪性肝疾患、2型糖尿病などの生活習慣病の改善という観点から、特に重要である。しかし、本邦で承認されている抗肥満薬は、そのリスクとベネフィットの観点から、日本人の肥満症患者のほとんどを占める軽度・中度の症例すべてに適応されるわけではない。したがって、現在も、安全性と有効性を兼ね備えた抗肥満薬が求められている。アブラナ科野菜に含まれるGRはもとより、BARDも既に他の疾患の治療薬として臨床で使われている薬物であり、安全性は高い。無論、さらなる検討を加える必要があるが、抗肥満薬として臨床応用できる可能性がある。
