Outline of Research at the Start

本研究では、熱産生機能を有する褐色脂肪細胞を誘導する低分子化合物を探索し、基礎代謝を亢進させる新規治療法の開発につなげることを目的としている。なぜなら、褐色脂肪細胞の数を活性化させて基礎代謝を上げることができれば、肥満症・メタボリックシンドロームやその関連疾患の発症・進展を抑制できるだけでなく、健康寿命の延長につながる可能性が考えられるからである。培養脂肪細胞を用いた検討から有望な化合物を選択し、マウスに投与することで肥満症の表現型が改善するか検証し、褐色脂肪細胞活性化薬の新規創薬につなげていきたいと考えている。

Outline of Final Research Achievements

A high-throughput screening of compounds targeting uncoupling protein 1 (UCP1) in adipocytes was conducted, and a compound targeting a G protein-coupled receptor that specifically promotes heat production in adipocytes was identified. After confirming that the compound had little cytotoxicity, the compound was administered to a high-fat diet-induced obesity model mouse. The compound administration induced weight loss and improvements in hepatic steatosis, glucose tolerance, and insulin resistance. Basal metabolism was also enhanced, and cold tolerance was also improved. Comprehensive gene expression analysis revealed a new mechanism for enhancing UCP1 expression via the p38MAPK signal pathway and its downstream pathways, Kcnc2 and Gm5627, leading to the acquisition of a patent.

Academic Significance and Societal Importance of the Research Achievements

本研究では化合物スクリーニングから、脂肪細胞に特異的に作用し、抗肥満効果を発揮する薬剤を同定できた。同定した化合物は脂質代謝や脂肪合成への影響は少なく、また、細胞毒性が強くないことから、安全で効果的な肥満症治療薬としての可能性が示された。この化合物の作用機序の探索から、これまで報告されていない複数の褐色脂肪細胞活性化機序が明らかになり、新たな肥満治療の標的分子となる可能性があると考えられた。
