Outline of Research at the Start

SOX10-VENUSマウスから新規ES細胞を樹立し、これを用いて基礎的なENS発生過程を経時的に観察、腸管神経幹細胞の誘導を行う。さらに、同細胞を用いた移植実験を予定する。同時にSOX10-VENUSマウスから作成した神経塊を用いてH病腸管部位の細胞環境の解析を行うことで、神経発達促進因子を探索し、幹細胞移植後の生着・発達を促進する物質をその後の移植実験でも検証することが可能となる。

Outline of Annual Research Achievements

本研究では、新規ES細胞を樹立することで未だ解明されていない腸管神経系の発生メカニズムの解析をもとにH病の病態解明を行い、さらには新しい治療リソースとして革新的な細胞治療方法の臨床応用に展開するための基礎研究を行う。SOX10-VENUSマウスから新規ES細胞を樹立し、これを用いて基礎的なENS発生過程を経時的に観察、腸管神経幹細胞の誘導を行う。さらに、同細胞を用いた移植実験を予定する。同時にSOX10-VENUSマウスから作成した神経塊を用いてH病腸管部位の細胞環境の解析を行うことで、神経発達促進因子を探索し、幹細胞移植後の生着・発達を促進する物質をその後の移植実験でも検証することが可能となる。今年度は、昨年新規に樹立したES細胞の腸管神経幹細胞への分化誘導方法確立に取り組んだ。さらにはモデルマウス腸管との共培養を行い、神経分化の過程を検証する。新規に樹立したSV-mES細胞から、ハンギングドロップ法でEB細胞を作製。3日目からSOX10発現が蛍光顕微鏡下で確認可能となった。これを浮遊培養に移行し、神経細胞塊としてDual Smad Inhibition法で効率的に分化誘導を行う。その後神経塊を作成し、正常およびモデルマウス腸管の小腸、結腸それぞれと共培養することで異なる分化傾向を示し、移植先の腸管における微小環境が腸管神経ネットワーク構築に重要な役割を果たしていることがわかった。

Reason

今年度は、ES由来腸管神経幹細胞より作成した神経塊をドナーとして用いた、正常およびヒルシュスプルング病マウス腸管とのin vitro共培養実験を実施した。それにより、正常マウス腸管との共培養では、神経塊から全周性に軸索を伸長し神経ネットワーク構築する様子が観察され、幹細胞の移動および分化も確認され、ドナー細胞としての可能性がみられた。一方で、ヒルシュルプルング病腸管との共培養では神経塊の大きさが小さく、軸索伸長がまったくみられなかった。このように、ドナーとしてのES由来腸管神経幹細胞の有用性および移植先の環境が神経発生には大きく関与していることがわかった。

Strategy for Future Research Activity

今年度はin vitro実験が進捗したため、今後は計画書にあるように、ドナー細胞の投与量、投与方法、投与時期を明確にしたのちにin vivo実験の実施を予定している。
