Outline of Research at the Start

多くの癌種では正常細胞と異なったメチル化修飾状態を持ち、癌の代謝/成長を支持している。メチル化修飾を受ける標的としてはDNA/RNAなどの核酸、ヒストンなどのタンパク質、ポリアミンなど多岐にわたる。ニコチンアミドはNicotinamide N-methyltransferase (NNMT)により不可逆的なメチル化修飾を受け、生成された1-methylnicotinamide (1-MNA)を細胞外に排出することで細胞内のメチル基レベルを調節している。本研究ではNNMTの治療標的としての可能性およびNNMT代謝物のバイオマーカーとしての有用性を明らかにする。

Outline of Annual Research Achievements

本研究課題の主題であるニコチンアミドメチル基転移酵素(NNMT)は申請者が報告した膵臓癌シングルセルアトラスから探索された分子であり、悪性細胞、腫瘍
関連線維芽細胞(CAF)などで発現が亢進することがmRNAレベルで確認できていた。

そのタンパク質発現をヒトすい臓がん病理標本100例を用いて解析を行ったところ、CAFについては一様の高発現が見られた一方で、悪性細胞においては症例間および症例内でも発現陽性細胞の頻度に大きな不均一性が見られた。またacinar-ductal metaplasia (ADM)の箇所でNNMT陽性細胞が増加していることが病理解析から観察され、シングルセルアトラスにおいてNNMT mRNA発現を再確認したところ、acinarから上皮細胞への分化移行部において発現獲得している様子が確認できた。上記のことは、acinar-ductal metaplasia(ADM)に伴う大きなエピジェネティック変化とメチル基転移反応のかかわりにNNMTが関与している可能性を想像させる結果である。

そこでマウス膵腺房細胞のex vivo培養によるADM誘導モデルを用いて、NNMTの発現抑制実験を行ったところ、ADMの抑制傾向が得られた。

臨床検体を用いた解析から、in silicoの解析、さらにはin vitroの実験に展開するようにウェットとドライの手法を活かして研究全体が滞りなく進んでいる。

Strategy for Future Research Activity

臨床検体の評価を増やしつつ、in vitroではCAF細胞や腫瘍細胞にも広げて、すい臓がんにおけるNNMTの包括的な役割を解明していく。
