慢性炎症の蓄積が加齢であるという”Inflammaging”は、超高齢化社会の課題であるフレイルや運動器不安定症を解決する大きなヒントであり、その開始点のdanger moleculesの自己由来分子群がDAMPsである。本研究ではDAMPsの一員HMGB1と機能的相補性が予想されるHMGB2に着目し、骨芽細胞特異的に成獣期でコンディショナル・ノックアウトしたマウスの骨を詳細に解析することで、HMGBファミリーの骨形成と骨リモデリングへの関わりを探究する。これによりHMGB1/2の加齢による骨代謝変化における意義と、inflammaging制御療法としての新規骨粗鬆症分子治療標的としての妥当性を検証する。

| Outline of Final Research Achievements |
In an increasingly aging society, the relationship between bone metabolism and low-grade chronic inflammation associated with aging(inflammaging) has emerged as a significant challenge in the treatment of osteoporosis. This study focuses on HMGB1 and HMGB2, which are known to trigger sterile inflammatory responses. The objective of the research was to investigate the specific roles of HMGB1 and HMGB2 in bone remodeling by using conditional knockout mice in which these genes are selectively deleted in osteoblasts. Although progress in the first year was delayed due to issues with mouse breeding, preliminary findings using a mouse model of osteoarthritis (OA) provided meaningful insights. HMGB1 appeared to be involved in early-stage bone loss. Moving forward, the study aims to deepen our understanding of HMGB1 and HMGB2 function in bone remodeling under inflammaging conditions. This will involve phenotype analyses in aged mice and in OA models.

| Academic Significance and Societal Importance of the Research Achievements |
Col1a1-Cre;Hmgb1 cKOマウスの解析で、若齢期では骨量変化は乏しかったが、OAモデルにおいてはcKOマウスで骨量減少が抑制され、Hmgb1が骨量減少に関与する可能性が示された。さらに、ラット肩関節を用いた新規肩OAモデル(mCTA)を確立し、Hmgb1の発現亢進とOA病態との関連を明らかにした。ゾレドロン酸によるCTAのOA変化抑制効果とHmgb1の骨における発現レベルが密接にリンクしていたことから、Hmgb1の機能抑制が骨量減少に対する有望な治療分子標的であることが示唆された。老齢マウスモデルを用いた今後の研究により、加齢炎症に伴う骨粗鬆症やOAの新たな機構解明が期待される。
