Outline of Research at the Start

本研究は、骨関節疾患の手術に使用するインプラントの感染耐性をあげ、感染の治療にも直接使用でき、かつ再生医療として骨再生能力の高い新規バイオインプラントを開発、臨床応用へとつなげることである。この開発により整形外科手術全般を劇的にかえ得ることになり、術後感染の撲滅、インプラント周囲の骨再生の促進により早期リハビリ、社会復帰など、高齢社会への健康寿命、歩行能力の維持への貢献が大いに期待される。

Outline of Final Research Achievements

We developed a novel bioimplant combining iodine-supported porous titanium and adipose-derived stem cells (ADSCs) to achieve both infection resistance and bone regeneration. The titanium was surface-treated and successfully loaded with iodine. ADSCs showed high viability and good adhesion to the surface, with uniform cell distribution in porous structures. Using PET mesh, we achieved even placement of spheroids on the implant. ALP mRNA expression confirmed osteogenic differentiation on the implant. A hybrid construct of collagen and ADSCs allowed for full three-dimensional coverage. Histological analysis demonstrated effective cell integration and tissue formation. This bioimplant offers promise as a regenerative material for orthopedic applications requiring both anti-infective properties and bone healing capacity.

Academic Significance and Societal Importance of the Research Achievements

高齢化に伴い増加する整形外科手術において、術後感染は重大な課題である。本研究では、感染抵抗性と骨再生能を併せ持つ新規バイオインプラントの開発を行った。ヨード担持チタンと脂肪由来幹細胞を組み合わせることで、生体親和性と抗菌性を両立した構造体を構築し、感染抑制と骨癒合促進の双方を実現した。幹細胞のスフェロイド化や全周接着法の導入により、三次元的な細胞配置と機能発現を可能としたことも特徴である。この技術は、再手術の回避や患者のQOL向上に貢献するだけでなく、再生医療と感染制御を統合した新たな治療戦略として、整形外科領域に革新をもたらす可能性がある。
