Outline of Research at the Start

前立腺癌は、癌細胞の特性だけでなく、腫瘍微小環境が癌細胞のさらなる悪性化や去勢抵抗性や化学療法、放射線治療の抵抗性獲得に関与するとされる。腫瘍微小環境は、癌細胞以外に線維芽細胞、脂肪細胞、免疫細胞などから構成される特有の環境である。近年、細胞膜にある不動性線毛である一次線毛が細胞増殖などに関与するシグナルハブであることが明らかになってきた。本研究では、前立腺癌微小環境を模した共培養系で、前立腺癌細胞、脂肪細胞、および相互作用における一次線毛の発現・機能解析を行い、各細胞および腫瘍微小環境における一次線毛の重要性を明らかにし、前立腺癌の診断・治療への新たな可能性を探ることを目的とする。

Outline of Final Research Achievements

In this study, we analyzed the expression of primary cilia in prostate cancer cells and mesenchymal cells by fluorescent staining. Primary cilia were detected in DU145 cells, but not in PC-3 cells, and was detected by changing cell adhesion conditions. On the other hand, primary cilia were detected in fibroblasts and adipocytes. The expression of primary cilia in the cabazitaxel-resistant lines suggested that there is a low relationship between the expression of primary cilia and the acquisition of drug resistance. However, the abnormalities of centrosome were observed. This study suggests that (1) primary cilia may be involved in carcinogenesis, (2) primary cilia expression is reversible, and (3) centrosome may affect the morphology or function of prostate cancer cells.

Academic Significance and Societal Importance of the Research Achievements

多くの薬剤が出てきたと言え、転移性去勢抵抗性前立腺癌の治療は大きな問題である。従来のホルモン関連の関係性でなく、根源的な細胞生物学に根ざした新しい治療標的につながる可能性がある。
