Outline of Research at the Start

再生医療の実用化に向けては、解決しなければいけない課題がある。目的とする臓器や組織の再生に最適な幹細胞の選別や幹細胞の培養は、様々な増殖因子や分化因子を様々な組み合わせて添加してする方法が主流であるが、添加因子の理想的組み合わせは未だ十分に決定されていない。
本研究では、これまでにわれわれが報告したiPS細胞の凍結切片上での培養法を応用して、どの幹細胞が目的臓器・組織の再生のための幹細胞として最適かを簡便かつ経済的に認識、選別できる新たな評価法の確立を目指す。

Outline of Final Research Achievements

In this study, the principal investigators applied the methods they had previously reported for the differentiation induction and quality control of iPS cells to culture various types of stem cells on frozen sections of the target organ to be regenerated. By using this method to evaluate the ease of differentiation into functional cells of the target organ, the aim of the study was to establish a new evaluation method that can easily and inexpensively verify what state of stem cells is optimal as a cell source for the regeneration of the target organ or tissue. As a result, when various types of human dental pulp stem cells were cultured on frozen sections of various mouse organs and tissues (brain, spinal cord, liver), it was found that the cell morphology approached that of the frozen sections.

Academic Significance and Societal Importance of the Research Achievements

再生医学において安定した組織再生のための細胞の供給源を確保することはきわめて重要と考えられ、ES細胞や成体幹細胞、iPS細胞を用いた再生研究が進められている。これまでに複数の成体幹細胞が報告されているが、それぞれがどの細胞に分化しやすいのかという方向での報告は国内外ともほとんどなされていない。今後、個別化再生医療を目指すにあたり、個々の患者において目的臓器を再生するにあたり、用いる幹細胞の最適化が簡便かつ安価に行えれば、より安全で経済的な個別化再生医療実現に向けて大きな一歩となるものと考えられ、本研究は次世代再生医療の実現に向けて大きな一助となる独創的かつ有意義な研究であると考えている。
