Outline of Research at the Start

組織幹細胞は単離時の特異性が不明確であることや培養による特性低下など臨床応用の課題となっている。臍帯由来幹細胞(UC-MSCs)は、細胞の調達、保管、および移植において、他の幹細胞源よりも有用であるとされている。しかし、UC-MSCsの機能的特性や分化指向性については未だに解明されていない。本研究では、scRNA-seq, scATAC-seq解析により、エピジェネティックなランドスケープ、それらの遺伝的類似性および機能的特性の評価を行うことで、UC-MSCsを構成する細胞と骨分化制御シグナルを同定し、このシグナル分子を活用したin vitro培養法を確立し、骨再生医療発展の一助とする。

Outline of Final Research Achievements

This study aimed to establish a foundation for regenerative medicine targeting cleft lip and palate (CL/P). Single-cell RNA sequencing (scRNA-seq) was used to compare umbilical cord-derived mesenchymal stem cells (UC-MSCs) from CL/P patients and normal UC, thereby elucidating differences in cellular properties and differentiation trajectories. The analysis identified a distinct cell population involved in osteogenesis, along with four candidate factors, some of which overlapped with CL/P-related genes previously reported in genome-wide association studies (GWAS). Experimental systems to evaluate the functional significance of these factors have been developed, and the findings are currently under journal review. Moving forward, we aim to validate these osteogenic factors using disease-specific iPS cells generated from surgical specimens, with the ultimate goal of applying the results to regenerative therapies.

Academic Significance and Societal Importance of the Research Achievements

本研究は、唇顎口蓋裂(CL/P)に関連する間葉系幹細胞の特性解析を通じて、先天性骨欠損の病態解明と再生医療への応用可能性を示した点で学術的意義が高い。特に、非侵襲的に採取可能で倫理的制約の少ない臍帯組織由来幹細胞を用い、一細胞遺伝子解析(scRNA-seq)およびゲノムワイド解析(GWAS)により骨形成に関与する新規因子と特異な細胞集団を同定したことは、幹細胞の分化制御機構の理解を深め、先天異常に対する新たな治療戦略の構築に寄与する社会的意義を有する。
