Outline of Research at the Start

申請者らは、う蝕病原性細菌のうち菌体表層にコラーゲン結合タンパクを発現しているミュータンスレンサ球菌(CBP 陽性株)が、感染性心内膜炎や脳出血などの全身疾患に関与していることを明らかにしてきた。一方で、これらの予防法については明確に確立できておらず、CBP 陽性株を口腔内に定着させないことが根本的な予防策といえる。本研究では、CBP 陽性株の口腔における定着部位、定着時期および定着メカニズムの特性や、CBP 陽性株を有する小児の口腔細菌叢の特性について明らかにすることで、CBP 陽性株の口腔内への定着を抑制する方法を確立するとともに口腔領域から全身疾患の予防法を提唱したいと考えている。

Outline of Final Research Achievements

Molecular biological analysis was performed to clarify the colonization environment of Streptococcus mutans bacteria expressing collagen-binding protein (CBP) on the surface (CBP-positive strains). Results of a survey of over 300 pairs of caregivers and related children showed mothers as the main source of infection for children with CBP-positive strains, with breastfeeding during infancy as one of the factors with effects on colonization of those strains in the oral cavity. In addition, analysis of breast milk components regarding their effects on S. mutans revealed an antibacterial effect of the enzyme lysozyme against those bacteria at concentrations normally found in breast milk. In contrast, lactoferrin, a glycoprotein, showed a slow bacteriostatic effect.

Academic Significance and Societal Importance of the Research Achievements

齲蝕病原性細菌であるミュータンスレンサ球菌のうち菌体表層にコラーゲン結合タンパクを発現している CBP陽性株は、感染性心内膜炎や脳内微小出血などの全身疾患に関与することが明らかになっている。本研究では、CBP 陽性株が口腔内の定着しやすい環境因子を明らかにするとともに、ミュータンスレンサ球菌に対する母乳中の抗菌成分の効果について明らかにした。本研究成果は、口腔内環境が全身の健康維持に重要であることを示すとともに、口腔内環境が形成される上での授乳習慣の重要性を提唱する一途となると考えられる。
