Outline of Research at the Start

最近、予備的実験より、火薬の燃焼エネルギーで駆動する新規の無針注入器(PJI)が、皮内へのDNA投与により、有針注射器より強力な抗腫瘍免疫を誘導することや、アジュバントを加えずに蛋白質投与のみでも、抗原特異的細胞性免疫を増強することも見出した。そこで、本研究では、モデル抗原および分子として卵白アルブミン(OVA)と免疫抑制性サイトカインIL-27を用い、OVAプラスミドDNA投与による抗腫瘍ワクチン効果、アジュバントフリーのOVA蛋白質投与による抗腫瘍ワクチン効果、さらには、IL-27プラスミドDNA投与による遺伝子治療への応用について、新規無針注入器の有効性とその作用機序を明らかにする。

Outline of Final Research Achievements

The current success of mRNA vaccines against COVID-19 has highlighted the effectiveness of mRNA and DNA vaccinations. To improve the efficiency and safety, we have investigated the potential application of a needle-free pyro-drive jet injector (PJI) for vaccination against cancers using ovalbumin (OVA) as a model antigen. Intradermal injection of OVA-expression plasmid into mice using the PJI, but not a needle syringe, rapidly and greatly augmented OVA-specific CD8+ T cell expansion in lymph node cells. OVA-specific in vivo killing of the target cells and OVA-specific antibody production of both the IgG2a and IgG1 antibody subclasses were greatly augmented. Intradermal injection of OVA-expression plasmid using the PJI showed stronger prophylactic effects against the progression of transplanted E.G7-OVA tumor cells. The intradermal injection possibly induced shear stress, whereby augmenting the uptake of the plasmid in dendritic cells and activating innate immune responses.

Academic Significance and Societal Importance of the Research Achievements

今日、ヒトワクチンに用いられているアジュバンドは主に水酸化アルミニウムだけであるが、アルミニウムは蓄積すると神経障害を誘導することがある。一方、mRNAワクチンも、塩基を修飾し炎症誘導能を抑えたmRNAの免疫原性を上げるため脂質ナノ粒子に内包し筋肉注射しているが、そのため、血流に入り全身を巡り炎症を誘導する可能性が指摘されている。このように化学的アジュバンドは副作用誘発の可能性が高いが、物理的アジュバンドは、一過性に作用するだけなので、局所貯留や全身循環の可能性がなく、副作用の心配が少なく、その上、PJIのワクチン効果は高いので有効なワクチン増強手段になる可能性が高く、この成果の意義は大きい。
