Outline of Research at the Start

猛威を振るう新型コロナウイルス感染症はSARS-CoV-2ウイルスによる空気感染(エアロゾル感染)が主要な感染経路とされている.この空気感染性ウイルスによる感染伝播の問題は,気道細胞上でのウイルス増殖から,ヒト-ヒト間,更に社会空間での感染動態を含む非常に複雑な問題である.この問題に対し,本研究では,細胞上でのウイルス増殖から社会スケールでの感染伝播までの多層スケールでのウイルス増殖・感染動態に着目し,全てのスケールを通底・支配する力学系数理構造を抽出し,多層スケールを接続可能な普遍的なウイルス増殖・感染ダイナミクスモデルの開発に取り組む.

Outline of Annual Research Achievements

呼吸器系の粘膜上皮に沈着したウイルス量を基にその後の増殖,生体防御反応を決定論的感染症モデルHost cell dynamics(宿主細胞,感染細胞,ウイルスRNA copiesと免疫因子の常微分方程式群)として定式化し,上気道粘膜内での3次元的なウイルス動態を明らかにする数理モデルを開発した.加えて,COVID-19のHuman challengeデータを基に数理モデルのモデルパラメータの最適化にも取り組んだ.これまで,鼻腔内でのウイルス動態に着目していたが,咽頭から気管にかけての気道領域におけるウイルス濃度予測にも展開した.加えて,感染者が咳をした際の突発噴流によって生成される咳飛沫の発生位置をオイラー液膜モデルを用いて,予測するとともに気道内粘液中でのウイルス濃度分布との統合により咳飛沫に含まれるウイルス濃度の予測も可能にしている.
さらに,室内環境中に在室している人々が接触し,咳などによって飛沫に暴露することを想定し,異なる対人距離での飛沫拡散および暴露解析を行った.様々な対人距離(0.7,0.9,1.1mなど)に加えて,感染者と非感染者の姿勢の違い(立位や座位)にも着目し,複数のケースでの曝露解析を実施し,感染防御のための適切な対人距離を明らかにした.
また,社会システム上での感染動態予測に対しては,年齢別の接触マトリックスをもとにした数理疫学モデルを構築し,年齢別の潜在的な感染リスクの予測を進めた.

特に大きな変更点や問題無く,当初の研究計画に従って研究を推進している.既に,本研究に関連する研究分野ではトップジャーナルとなるComputer Methods and Programs in BiomedicineならびにPLOS Computational Biolodyなどに複数の研究成果も公表されており,順調に研究が進展していると判断している.

Strategy for Future Research Activity

今後は,人間でのウイルス感染に限らず,サルなどの実験動物に数理モデルを適用し,人間との比較を行ったうえで,スケーリング則を検討する.更に咳によって生成された飛沫の輸送挙動の高精度化に向けて,詳細な口腔形状を持つ数値人体モデルでの飛沫拡散解析を行い飛沫暴露予測を実施していく.
