Outline of Research at the Start

細胞の老化は、臓器の機能低下を引き起こし、様々な疾患のリスクを上昇させる。近年、老化に伴う細胞レベルでのエピゲノム変化の蓄積が、病的な加齢性変化の起因となることが明らかになった。研究者は、これまで炎症性サイトカインを多量に産生する病原性ヘルパーT細胞が、老化マウスで特異的に出現することを見出した。そこで、本研究は、老化に伴うエピゲノム変化を“エピゲノム老化”として着目し、研究者が同定した病原性老化ヘルパーT細胞を用いた1細胞レベルでの網羅的エピゲノム・遺伝子発現量解析、超解像度顕微鏡による先端イメージング解析を通じて、エピゲノム老化による老化形質獲得形成機構の解明を目指す。

Outline of Final Research Achievements

In aged mice, Aspergillus-induced lung inflammation was exacerbated, accompanied by marked fibrosis in the alveolar regions. Single-cell RNA-seq analysis identified cytotoxic Cytotoxic CD4 T cells that specifically emerged in aged mice. These cells exhibited features of epigenomic aging, including significantly reduced expression of the histone-modifying enzyme Ezh2 and decreased H3K27me3 levels. Furthermore, Ezh2 suppressed pro-inflammatory cytokine production via downstream transcription factors, and its dysfunction contributed to chronic inflammation and fibrosis. These findings suggest that age-associated epigenomic changes induce pathogenic traits in Cytotoxic CD4 T cells, potentially promoting the progression of age-related diseases.

本研究は、加齢に伴うT細胞のエピゲノム変化、特にEzh2の発現低下がCytotoxic CD4陽性T細胞の異常な分化と機能獲得を誘導し、慢性炎症や線維化を促進することを明らかにした。これは、加齢に伴う免疫異常の分子基盤を明確化するものであり、エピゲノム老化と病原性T細胞の関連を初めて示した重要な成果である。さらに、本研究は老化免疫の分野における新たなパラダイムを提供し、加齢関連疾患の病態解明と治療標的の探索に貢献する。
