Outline of Research at the Start

樹状細胞(DC)は、免疫応答の司令塔的な役割を担う重要な免疫細胞である。特に、2型古典的DC(cDC2)はヘルパーT細胞の誘導を行う一方で、短命な細胞であることが知られている。このcDCの恒常性は厳密に制御されていると考えられるが、その詳細な分子機序は未だ明らかになっていない。そこで本研究では、cDC2に高度に発現する膜型分子であるSIRPαとそのリガンドであるCD47を介したcDC2の生存制御機構について焦点を絞り検討を行う。本研究の成果は、DCの生存制御機構のより深い理解と、自己免疫疾患などの病態解明と新たな治療法開発へとつながることが期待される。

Outline of Final Research Achievements

In this research, we focused on the involvement of CD47-SIRPα system in the regulation of dendritic cell (DC) survival by using tomoxifen-inducible SIRPα knockout (iKO) mice. RNA-seq of sorted DCs from SIRPα iKO mice revealed that expression of cell death associated genes such as gene N was upregulated in the absence of SIRPα in DCs. To elucidate the role of gene N in the surivival of SIRPα-deficient DCs in vivo, we generated SIRPα/N double KO (DKO) mice. We demostrated that the number of DCs is recovered in the DKO mice compared with SIRPα KO mice. Thus, our findings indicate that SIPRα regulate DC survival by inhibiting gene N upregulation.

Academic Significance and Societal Importance of the Research Achievements

樹状細胞は免疫応答を駆動する上で最初のステップを担う重要な免疫細胞であり、その恒常性は厳密に制御されていると考えられている。一方で、その生存制御機構についてはいまだに明らかになっていない点が多い。本研究では、樹状細胞(DC)の生存制御機構の分子機序について焦点を当てて研究を行い、DCの生存がSIRPαおよび因子Nを介して制御されていることを明らかにした。さらに、SIRPαおよび因子NによるDCの生存制御機構が自己免疫疾患の発症においても重要な役割を果たすことが明らかとなった。DCの恒常性維持機構と免疫システム全体に関する理解を深め、自己免疫疾患の病態解明に寄与することができた。
