Outline of Research at the Start

全身性強皮症は全身性自己免疫疾患で、本邦における指定難病であり、有効な治療法はない。近年、この病態形成には免疫系の主要構成因子であるB細胞、中でも自己抗体産生の担い手である自己抗原反応性B細胞が全身性強皮症の病態に重要な役割を果たすことが想定されるが、数は少なく、直接的な病原性の解析法が十分に確立していない。今回の研究では、医工連携研究によって開発された少数細胞のタンパク質レベルでの解析システムを用い、全身性強皮症における自己抗原反応性B細胞の役割を検討し、広く自己免疫疾患における自己抗原反応性B細胞の役割の理解を深め、新たな治療ターゲットの同定と治療法開発へとつなげることを目指す。

Outline of Annual Research Achievements

本研究では、マイクロ空間を用いた独自の技術と方法論を用い、全身性強皮症(systemic sclerosis; SSc)における自己反応性B細胞の機能と役割を明らかにす
ることを目的とした。まず抗Topoisomerase(topo) I抗体陽性SSc患者における末梢血中のtopoI特異的B細胞を抽出し、サイトカイン産生能などの解析を行った。
様々なサイトカインのうち、Interleukin (IL)-6やIL-23などの炎症性サイトカインを産生するB細胞やIL-10やIL-35などの抑制性サイトカインを産生するB細胞
がtopo I特異的B細胞集団中に存在することを見出した。これらの細胞内シグナル伝達経路やサイトカインがB細胞に与える機能解析を行い、それぞれの細胞集団に特徴的な経路が見られた。
次に強皮症モデルマウス解析に関して、topoI誘発SScモデルマウスを用いた。B細胞のaffinityの上昇とサイトカイン産生能の関係について検討を行った。topo I蛋白を免疫する毎にB細胞のtopo Iに対するaffinityは強められた。low affinityなB細胞が多いモデルでは、抑制性のサイトカインを産生するB細胞の割合が多く、high affinityなB細胞が多いモデルでは、炎症性サイトカインを産生する割合が多かった。
コンディショナルノックアウトマウスを用い、特定のサイトカインが強皮症の病態形成に与える影響について検討を行い、ある特定のサイトカインが強皮症の病態形成に重要であることが示唆された。
