Outline of Research at the Start

これまで我々は、紫外線により皮膚で誘導されるAhRの一つであるFICZが、ヒト表皮ケラチノサイトからのIL-17関連因子を増幅し、上皮系皮膚悪性腫瘍の進展に関与することを明らかにしてきた。IL-17は皮膚の抗菌ペプチドであるLL37の樹状細胞やマクロファージなど単球由来細胞で上昇すること、LL37は抗原提示細胞における自己抗原の取り込みを増加することが知られている。以上の観点から、本研究は早期と進行期CTCLにおける紫外線の役割を、AhRリガンドにより誘導される免疫反応をTAMs中心に解析し、紫外線が早期および進行期CTCL進行に与える影響をTAMs中心に解析することを目的とする。

Outline of Final Research Achievements

The effect of a substance induced from keratinocytes by ultraviolet light (FICZ) on tumor cells and tumor-associated macrophages (TAMs) in cutaneous T-cell lymphoma (CTCL) was examined. The effect of PAI-1 on TAMs was examined, and PAI-1 increased the production of MMP-9. Furthermore, PAI-1 increased MMP-9 production in CTCL cell lines, confirming that suppression of MMP-9 in the mouse CTCL model reduced vascular endothelial cells in the tumor.

Academic Significance and Societal Importance of the Research Achievements

本研究ではPAI-1の抑制がCTCLにおいて重要な役割を果たすことが示され、CTCLに対してPAI-1阻害薬が有効である可能性を示された。将来的にはCTCLの治療薬としてPAI-1阻害薬の適応拡大が進められることが期待される。以上、本研究は研究期間中に新たなCTCLの生物学的メカニズムを解明したことに加え、近い将来の新規CTCL治療薬の開発につながる画期的な研究であったと考える。
