Outline of Research at the Start

難治性造血器腫瘍である多発性骨髄腫は, 過剰に産生されるM蛋白により, 腫瘍細胞では小胞体ストレス応答(UPR)が恒常的に活性化している. UPRにおいて, 中心的役割を担うIRE1αの活性化にABLが関与する. 申請者はABL-IRE1α経路に着目し, 骨髄腫細胞株と, 患者由来の腫瘍細胞を用いたin vitroの実験, xenograft mouse modelを用いたin vivoでの実験により, ABLがIRE1α経路, UPRに及ぼす影響を明らかにし, 骨髄腫におけるABL-IRE1α経路の新規治療標的としての可能性について検討する.

Outline of Annual Research Achievements

難治性造血器腫瘍である多発性骨髄腫は, 過剰に産生されるM蛋白により, 腫瘍細胞では小胞体ストレス応答(UPR)が恒常的に活性化している. URPは細胞の生存・恒常性維持に重要であるが, 過剰な小胞体ストレス下では細胞死を誘導することが知られている. UPRにおいて, 中心的役割を担うIRE1αの活性化にABLが関与しており, 申請者はABL-IRE1α経路に着目した. 本研究課題では, ABLがIRE1α経路, UPRに及ぼす影響を明らかにし, 骨髄腫におけるABL-IRE1α経路の新規治療標的としての可能性について検討している. 本研究課題では, ABL阻害剤は, ABLの構造変化を誘導するアロステリックABL阻害剤であるGNF-2とasciminibを主に使用して解析を進めており, いくつかの結果を得ることができた. 1)ヒト骨髄腫細胞株と患者由来骨髄腫細胞株は, ABL阻害剤によってIRE1α経路が活性化し, 細胞死が誘導されることを明らかにした. 2) ABL阻害剤であるasciminibは, 細胞質に存在するABLを小胞体へと局在変化させることを明らかにした. 3) RNAシーケンス, qPCR法によりasciminibはIRE1α経路だけでなく, UPRにおける広範な経路を活性化させることを明らかにした. 4) asciminibはアポトーシス誘導因子であるCHOPの発現を誘導することを示し, IRE1α阻害剤を併用するとCHOPの誘導が増強され, 抗骨髄腫効果が増強されることを明らかにした.
本研究課題の結果より, アロステリックABL阻害剤であるGNF-2やasciminibがABL-IRE1α経路を標的として, UPRの過剰な活性化・UPR制御機構の変容という観点から, 多発性骨髄腫の新規治療薬になり得る可能性が示唆される.

Strategy for Future Research Activity

ヒト骨髄腫細胞株と患者由来骨髄腫細胞を用いたABL阻害剤の生物学的効果の評価をさらに検討し, in vivoでの実験系も組み合わせ, 骨髄腫におけるABL-IRE1α経路の新規治療標的としての可能性について検討する.
