Outline of Research at the Start

膵臓がんは他のがんに比べて予後が著しく悪いため、様々な角度から治療法の研究が試みられています。がんは自身が生き残るためにオートファジーと呼ばれる再生機能を駆使しています。我々はがん細胞内のエネルギー産生を行う小器官であるミトコンドリアの代謝とオートファジーの関連に着目しました。
本研究では、動物実験で膵臓がんのオートファジーに関わるミトコンドリア関連遺伝子を発見し、それを標的とした新規治療法の開発が可能かどうかを検証していきます。

Outline of Final Research Achievements

This study investigated the role of acid β-glucosidase (GBA), a lysosomal enzyme involved in sphingolipid metabolism, in pancreatic ductal adenocarcinoma (PDAC). GBA deficiency is linked to mitochondrial dysfunction. The researchers knocked down GBA in human PDAC cell lines (PANC-1, BxPC-3, and AsPC-1) using siRNA and analyzed various factors such as GBA enzyme activity, cell viability, reactive oxygen species (ROS) generation, and mitochondrial health. Results showed that GBA knockdown reduced cell proliferation, induced apoptosis, and impaired mitochondrial function, particularly in PANC-1 and BxPC-3 cells. Additionally, the combination of GBA suppression and the chemotherapy drug gemcitabine had a stronger antiproliferative effect than gemcitabine alone. Overall, the study suggests that GBA suppression could be a promising therapeutic approach in PDAC treatment due to its significant antitumor effects.

Academic Significance and Societal Importance of the Research Achievements

本研究において、膵癌におけるライソゾーム酵素群のひとつであるGBAの機能を解析し、抗癌剤耐性のメカニズムを明らかにするとともに、膵癌治療における基礎的知見を提供した。本研究の意義は、膵癌におけるミトコンドリアダイナミクスに関わる分子メカニズムを世界に先駆けて解明し、治療へ応用を目指す点である。
