Outline of Research at the Start

脳は多彩で豊富な脂質を含み、その量的・質的な変化は、脳機能に影響をおよぼすことが知られている。申請者は、脂肪酸伸長酵素Elovl6の欠損により、空間学習や恐怖記憶の障害が認められること、樹状突起スパインの形態に異常をきたすことを見出している。本研究では、Elovl6による脂肪酸組成の制御が、樹状突起スパインの形態や神経回路網の形成を制御するメカニズムを分子レベルで明らかにし、アルツハイマー型認知症やパーキンソン病のような神経変性疾患の新規予防法・治療法の開発に貢献することを目指す。

Outline of Final Research Achievements

Analysis of neuron-specific Elovl6 null mice revealed abnormal dendritic spine morphology, but no significant changes in neurogenesis or neuron-astrocyte interactions were observed in brain-specific Elovl6 null mice. Spatial learning and memory were also impaired, but to a lesser extent than those in brain-specific Elovl6-deficient mice. There was also a significant decrease in Elovl6 expression in the brains of patients with AD, suggesting the importance of Elovl6 in the nervous system.

Academic Significance and Societal Importance of the Research Achievements

現代社会では精神・神経疾患の患者数が爆発的に増えており、その予防・治療法の確立は急務である。これまでにもそれらの疾患に対する治療薬は開発されているものの、大きな副作用が生じる可能性があること、さらに、根治が困難であることが課題として残っている。本研究で着目している脂質は脳に豊富に含まれており、その量的・質的な制御は機能維持に重要であることが知られている。Elovl6による脂肪酸組成の制御は、脳・神経の研究における新しい概念の提唱とともに、特定の神経を標的としない予防・治療法の開発に有用である可能性が示唆され、その社会的意義は大きいと考える。
