Outline of Research at the Start

エンベロープウイルスの出芽機構を利用した自己集合型蛋白質ナノ粒子の被膜化技術、細胞質への蛋白質ナノ粒子の送達技術の開発を進め、SARS-CoV-2抗原を持つ被膜蛋白質ナノ粒子 (EPN: enveloped protein nanoparticle) のCTL誘導型ワクチンへの応用を目指す。ナノ粒子に付与する被膜化モジュールの改良を行いEPN産生の効率化を進める共に、ナノ粒子の細胞質送達率の向上を目指す。また、経鼻免疫やアジュバンドの改良を進め、細胞性免疫のみならず粘膜免疫誘導型のワクチンの開発を進める。また、ハムスターを用いたウイルス感染実験によりワクチン効果について検証を進める。

Outline of Final Research Achievements

This study aimed to develop a next-generation COVID-19 vaccine with improved safety, addressing issues associated with existing vaccines (such as mRNA vaccines), including adverse reactions and insufficient induction of cytotoxic T lymphocytes (CTLs). Specifically, a novel envelope protein nanoparticle (EPN) vaccine was developed by using Japanese encephalitis virus-like particles (JEV-VLPs) as a carrier and conjugating them with the SARS-CoV-2 spike receptor-binding domain (S-RBD) antigen. In mouse immunization studies, the vaccine induced strong neutralizing antibody and cellular (Th1-type) immune responses even at low antigen doses, and demonstrated cross-neutralizing activity against SARS-CoV-2 variants such as Alpha, Beta, Delta, and Omicron. This vaccine holds promise as a next-generation vaccine with both high safety and efficacy.

Academic Significance and Societal Importance of the Research Achievements

本研究によって、JEV-VLPをキャリアとした新規EPNワクチンが、低い抗原量で高い中和抗体と細胞性免疫(Th1応答)を誘導する基盤技術が確立された。また、SARS-CoV-2変異株(Alpha, Beta, Delta, Omicron等)に対しても交差中和活性があることが示された。既存のmRNAワクチン等が持つ重篤な副反応課題を克服し、安全性と有効性を両立した次世代型蛋白質ワクチンの可能性を示した。これにより、将来のパンデミックや複数回接種の必要性に対し、より安全で効果的な解決策を提供することが期待される。
