Outline of Research at the Start

Outline of Final Research Achievements
Topical application of the mTORC1 inhibitor sirolimus, used for treating skin lesions in tuberous sclerosis complex (TSC) patients, improved and resolved seizures in a patient without increasing blood sirolimus levels. This suggested a novel blood-independent pathway for sirolimus transport from the skin to the brain. Therefore, we investigated this novel mechanism of drug delivery from the skin to the brain. In a TSC model mouse, topical application of sirolimus improved seizures. However, when the innervating nerves at the application site were transected, this effect was lost, and the sirolimus concentration in the brain was reduced to zero. These findings confirmed that sirolimus was delivered to the brain and exerted its effects via a novel, nerve-mediated skin-to-brain pathway.

Academic Significance and Societal Importance of the Research Achievements
