Outline of Research at the Start

本研究は,古環境学と微生物学の融合を通して,過去の地球環境の転換点(tipping point)に応答して変化する海底堆積物の岩相が,同時に現在の海底下生命圏の分布や群集組成を規定するという仮説を検証する.この仮説はこれまで浅部海底下研究で示唆されている.本研究では,さらに深堀りするため,過去の地球環境の転換点で形成された多様な地質学的境界(ハイエイタスや明暗色互層など)に伴う微生物群集の構造変化と分布を調査する.

Outline of Annual Research Achievements

本研究課題は,古環境学と微生物学を融合させる新たな学術体系の開拓を通して,過去の地球環境の転換点(ティッピングポイント)が現在の海底下生命圏を規定しているという仮説の検証に挑む.海底下深部に広がる地球最大の微生物生命圏は,生命生存の極限的に低いエネルギー条件であるにもかかわらず,1億年前に堆積した地層においても生きた生命を宿す環境として知られている.泥質堆積物中で棲息する微生物は孔隙サイズが小さいため,海底堆積物を移動できない.この海底堆積物の岩相,つまり現在の海底下生命の生息環境としての特性は,過去の気候や海洋環境変化に応答して変動する.過去の環境変動が現在の海底下微生物の群集構造・分布を少なくとも部分的に規定していることは,いくつかの浅部海底下研究から示唆されてきた.そこで本研究では,過去の地球環境の大きな転換点で形成された地質学的境界1)ハイエイタス,2)火山灰層,3)明暗色互層,4)生痕化石に注目し,その地質学的境界に伴う「生の」微生物の群集構造の変化と分布を調査する.1)ハイエイタスは,長期的間隙が作る境界である.ハイエイタスの上下での現生微生物の群集構造や微生物細胞数の変化を追う.2)火山灰層は高間隙率で透水性が高いため,上下位の泥質堆積物と強い物性のコントラストを生む.火山灰層とその上下位の堆積物を分析し,噴火前・中・後の堆積物の微生物群集構造を解析する.3)日本海の明暗色互層は,酸化還元条件のコントラストを持つ地質学的境界である.これは氷期-間氷期の海水準変動に呼応している.日本海の明暗色互層を回収し,酸化還元変動に応じた微生物群集組成を解析する.4)生痕化石は堆積後に底生動物が作る複雑な形態の地質境界である.生痕化石の中と外,そして海底下浅部から深部にかけて微生物群集構造や分布がどう変化するのかを詳らかにする.

2: Research has progressed on the whole more than it was originally planned.

Reason

2022年に白鳳丸KH-22-3次研究航海で沖縄トラフの採泥が実施され,広域テフラ(鬼界カルデラ7 ka, 桜島13 ka)を含む海底堆積物が採取された.また,2023年度には新青丸KS-23-5次研究航海で日本海秋田沖で採泥が実施され,明暗色互層と火山灰層(大山草谷原21 ka)の回収に成功した.2024年度には,太平洋シャツキー海台で火山灰層やハイエイタス,生痕化石を含む堆積物が回収され,これで本研究でターゲットとした4つの地質境界「明暗互層」「火山灰層」「ハイエイタス」「生痕化石の内外」の全てのセットが揃った.ただし,ハイエイタスについては,その時間間隙を推定するための年代層序学的検討が必要である.

2024年度には,白鳳丸KH-24-1次研究航海(2024年4月2~19日)が実施され,北西太平洋シャツキー海台でピストンコアラーで遠洋性堆積物を採取した.本研究課題のターゲットである火山灰層を含む地層,生痕化石を豊富に含む堆積物,さらにハイエイタスが疑われる層準が回収できた.これらの試料は船上で半割し岩相記載を行った.航海後にコア試料は全て高知コア国際研究所に輸送され,マルチセンサーコアロガーを用いて密度や帯磁率など物性を測定したほか,X線CTスキャンによる内部構造の観察を行った,火山灰層,ハイエイタスの周辺と生痕化石の内外を判定し,微生物分析用にサンプリングした.現在,微生物分析を行っている.

Strategy for Future Research Activity

上記の通り,これまでに白鳳丸KH-22-7次研究航海(沖縄トラフ,奄美北西沖),KH-24-1次研究航海(シャツキー海台),新青丸KS-23-7次研究航海(日本海)で実施されたピストンコア試料(堆積物コア)で,ターゲットである4つの地質境界(明暗互層,火山灰層,ハイエイタス,生痕化石の内外)をほぼ全て入手できた.最終年度にあたる2025年度は,これらのコア試料の年代層序学的検討,古環境学的検討,火山灰層の同定,そして微生物学的検討を行う.微生物分析用の資料はすでに分取して冷凍保管している.生痕化石の内外における微生物細胞数や群集組成の研究には,その冷凍保管試料から,局所的なサンプリングを行う必要があるが,その手法の開発が必要である.開発と並行して進める.また,シャツキー海台で採取されたコアに含まれるハイエイタスらしき構造について,どの程度の時間間隙があるのかを,年代層序学的検討により明らかにする.
最終的に,これらの結果を統合して,各地層(岩相)境界において,どのように微生物細胞数と群集組成が規定されているかを明らかにし,本研究課題の最も重要な科学的命題である,過去の地球のティッピングポイントが,どのように海洋環境を変え,その結果として起こる岩相の変化(地質境界)が現在生息する微生物の群集組成や分布をどのように規定しているか,について検討していく.
