Outline of Research at the Start

乾燥や凍結により新陳代謝が完全に停止しても、その後、水分の補充や氷の融解により生命活動を再開する脅威的な耐乾燥性、耐凍結性を示す生物がいる。これらの生態は「cryptobiosis」として知られ、糖類のガラス転移が関与していると考えらている。糖類は、水の代替物質として作用しつつ、生体組織をガラス状態に封入し、周囲の劣悪な環境から長期間にわたり保護するとされる。本研究では、フェムト秒時間分解過渡吸収スペクトル測定等の超高速分光法により、糖ガラス中でプロトン・電子移動、光異性化等の化学反応ダイナミクスがどのように抑制されているのか、実時間測定することによりその驚異的な保護機能の解明を行う。

Outline of Final Research Achievements

In order to elucidate the suppression of molecular motion by the saccharide glass transition, we conducted experiments utilizing fluorescence red-edge effect (REE) and femtosecond transient absorption (TA) spectroscopy. We measured the REE of Auramine O (AuO) in saccharide glasses and observed the suppression of torsional motion of the phenyl groups. When femtosecond TA spectroscopy was performed on a similar non-fluorescent dye, phenol blue (PhB), it was found that non-radiative decay was not suppressed even when it was encapsulated in polymer glass, suggesting that the non-radiative decay of PhB does not involve large scale molecular motion. In addition, in order to apply these techniques to biological samples such as proteins, preliminary TA spectra measurements were performed on the light-harvesting antenna complex, LH2, of the purple photosynthetic bacterium.

Academic Significance and Societal Importance of the Research Achievements

生物の中には「クリプトビオシス」と呼ばれる驚異的な耐乾燥性や耐凍結性を示すものがある。たとえば、アフリカの砂漠に住むネムリユスリカの幼虫は、新陳代謝が完全に静止した17年間の乾燥状態の後に水を加えると蘇生したとの報告がある。クリプトビオシスの際、糖やその他の物質がガラス転移を起こし、極度に分子運動が抑制され、生体物質の劣化に関与する不要な化学反応を防止すると考えられる。そのメカニズムを解明することは学術的な意義のみではなく、臓器や生体物質の長期保存等の医学的な応用が期待できる研究であり、社会的な意義も大きい。そこで我々は、糖のガラス転移に関して分光学的な実験手法による研究を行ってきた。
