Outline of Research at the Start

真核生物では一般に1本のmRNA上に1つの読み枠(open reading frame: ORF)が存在し、1つのタンパク質が翻訳されると考えられてきた。しかし、近年のゲノム科学的研究により全体の35-50%ものmRNAにおいて実際に発現するORFの上流に数ペプチドをコードする小さなRNA(upstream ORF: uORF)が存在し、下流のORFの発現を制御していることが示唆されてきたが、その制御機構については完全に解明されていない。この課題を解決するため、本研究では植物RNAウイルスを利用して遺伝子発現制御機構の解析を行う。

Outline of Final Research Achievements

In this study, we analyzed the mechanism of gene expression through leaky scanning usging plant viruses. 5' untranslated region (5'UTR) of sgRNAs encoded by viruses of the genera Potexvirus, Lolavirus and Carlavirus was found to be significantly short and we named short UTR (sUTR). Several lines of analyses on the function of the sUTR revealed that the length of the 5' UTR acts as a regulator of leaky scanning, similar to the known Kozak sequence, and that the length of the sUTR is optimized for viral infection by regulating and balancing the expression levels of downstream viral genes.

Academic Significance and Societal Importance of the Research Achievements

真核生物のmRNAの大部分はuORFを持つことが明らかにされており、その制御機構の大部分はリーキースキャニングによると考えられている。本研究ではsUTRの長さが従来のKozak配列による制御に加えて、新たなリーキースキャニング制御機構であることを明らかにした。現状uORFがコードするペプチドが機能を持つかどうかを予想する術はないが、本研究で5'UTR が長いほど発現量が上がることが明らかになったため、5'UTRが長いuORFに絞り込むことによって機能的uORFの探索を容易にすることが可能になると予想される。このように本研究の成果はあらゆる生物学分野に幅広い波及効果がある。
