This study aimed at elucidating an effector-independent mechanism that promotes proliferation of the phytopathogenic bacterium Pseudomonas syringae under high humidity conditions. A screening of approximately 1,600 transposon mutants identified 35 mutants with reduced bacterial growth in plants under high humidity. Further analysis of these mutants revealed that several genes involved in amino acid biosynthesis are dispensable for growth in nutrient-rich media but are essential for growth in plants under high humidity. In addition, a two-component system was identified as being transcriptionally activated under high humidity to promote bacterial proliferation by switching gene expression patterns from effector-mediated immune suppression to nutrient acquisition.
