Outline of Research at the Start

「マイナー(U12)イントロン」は99%以上を占めるメジャー(U2)イントロンとは対照的に、全イントロンのわずか0.3%でありながら細胞の生存や恒常性の維持に不可欠な遺伝子に進化的に保存されている。本研究では、進化学・遺伝学・腫瘍学の観点から、マイナーイントロンの存在意義、それらが制御する遺伝子発現調節機構、その異常がもたらす発癌への未知の作用を先進的な手法を用いて解明していく。

Outline of Final Research Achievements

Only 0.3% of all introns have characteristic evolutionarily conserved branch point or 5′ sequences, and interestingly, they are usually found in genes essential for cell survival or homeostasis. Such specific introns are called "minor introns (U12 type)" due to their small number, and are removed by a different mechanism than normal introns. In this study, we explored the relationship between minor introns and cancer and the significance of their evolutionary conservation.

Academic Significance and Societal Importance of the Research Achievements

マイナーイントロンの制御破綻は血液がんを中心に認められているが、そのような特殊なスプライシング機構がなぜ進化の過程で保存されてきたのかは全く明らかになっていなかった。そこで、Pten遺伝子のマイナーイントロンをゲノムから除去したマウスモデルを作成し、造血幹細胞の機能を評価したところ、ストレス下において幹細胞性の維持が困難となることが明らかとなり、進化的保存の意義の一端を明らかとした。この研究はマイナーイントロンの制御破綻と発がんや老化をつなぐ上で、土台となる成果と言える。
