自閉症を含む発達障害の発症につながる分子メカニズムには不明な点が多い。私たちの研究グループは、発達期の臓器に際立って豊富に発現する遺伝子として「若年性遺伝子 (juvenility-associated genes, JAG)」を同定した。JAGの中には機能未知なものがたくさん含まれたが、本研究で着眼する遺伝子は脂質の代謝に関わることが発見された。さらに同遺伝子の変異が発達障害の発症に関連することが示唆された。本研究では同遺伝子の分子機能を詳細に明らかにするとともに発達障害の発症につながるメカニズムを解明し、新規の治療戦略を確立することを目的とする。

| Outline of Final Research Achievements |
Cells sustain life by active molecular metabolism, yet its full picture remains elusive. In this study, we focused on a molecule whose nuclear envelope translocation is associated with changes in cellular phospholipid levels. Functional analysis revealed that this molecule translocates to the nuclear pore complex (NPC) in response to lysophosphatidic acid depletion and suppresses total phospholipid levels. Mechanistically, the molecule was suggested to target and inhibit a phospholipid-synthesizing enzyme that also localizes to the NPC. These findings identify a previously uncharacterized set of molecules responsible for regulating the amount of nuclear envelope membrane, thereby advancing our understanding of the molecular basis of cellular metabolism.

| Academic Significance and Societal Importance of the Research Achievements |
脂質代謝は一般に小胞体にて起こるとされている。しかし、実際には核膜にも脂質代謝調節分子の局在があり、核膜が脂質代謝の場となる知見が徐々に蓄積されている。我々の研究からも、小胞体だけでなく核膜においても脂質やリン脂質の合成が起こり、それらの機序が神経発達症などの疾患とも関連することが明らかとなった。これらの知見は細胞代謝の理解に一石を投じ、実像の理解に一歩前進するものである。社会的意義としては、医学的な観点から、本計画で同定された機構は神経発達症 (neurodevelopmental disorder)の患者で変容していることが国際共同研究で明らかとなり、治療開発につながる可能性がある。
