本研究で、低転移性がん亜集団から放出されるエクソソームが高転移性がん亜集団の幹細胞性・転移を促進することを示す。本研究を通して、エクソソーム内のがん促進因子を同定する。体液中の促進因子の測定は、乳がん再発・転移の予測を可能にする。促進因子の抑制剤投与は新たな再発予防に繋がる。促進因子のインヒビターを入れた人工エクソソームによる新規乳がん治療法の開発を目指す。予後不良(10年無再発生存率50-60%)のTNBC乳がんの標準療法は確立していない。本研究は、エクソソームに着目し、乳がんの転移・予後予測、新規治療法開発を目指す世界初の試みであり、挑戦的研究として意義は極めて大きい。

| Outline of Final Research Achievements |
The role of exosomes in cancer metastasis was clarified as follows. 1.:There are two types of 4T1 cells, low metastatic and highly metastasized, and the metastasis of highly metastatic cancer cells is further promoted by Wnt7a contained in exosomes released from low-metastatic cancer cells. Wnt7a acts on tumor angiogenesis and forms an environment for cancer growth. 2: Administration of miR-200b-3p-rich exosomes into the tumors of tumor-bearing mice suppresses angiogenesis and suppresses tumor growth. 3: Exosome release from cancer cells and T cells is regulated by SPRRED2, and the ability of cancer cells to proliferate and invade is altered through exosome release by regulating the expression of SPRED2.

| Academic Significance and Societal Importance of the Research Achievements |
①がん細胞のサブクローン間のコミュニケーションが、がんの進行に重要な役割を果たしていることを示した。低転移性がん細胞はエクソソーム中のWnt7aを介して高転移性がん細胞の転移能を増強する能力を持つことを初めて証明し、Wnt7aがTNBC患者の治療の標的となる可能性を示した点は意義がある。②標的因子を含むエクソソームにより治療戦略が可能であることを示した。③腫瘍環境内のコミュニケーションを担うエクソソームは、SPRED2により調整されることを示した。SPRED2を標的にしたエクソソーム治療の可能性が示唆された。
