Outline of Research at the Start

GMの構成変化による細菌群の代謝変化がもたらす宿主における代謝異常とそれに伴う各種生活習慣病の発症には、特定の菌種単独による代謝作用だけでなく、GM間での代謝ネットワークを介した相互代謝作用が主に関与すること、そしてGM中の複数の細菌種の相互作用を明らかにすることで、各種生活習慣病の発症ならびに症状悪化リスク評価が可能となることを実証し、上記関連性を明確にするフレームワークの開発を目指す。

Outline of Final Research Achievements

It has been pointed out that there is a relationship between various systemic diseases, including lifestyle-related diseases, and intestinal dysbiosis. In this study, we aimed to clarify whether intestinal dysbiosis actually leads to an increased risk of developing various systemic diseases, and if so, what kind of diseases and what kind of dysbiosis characteristics are shown, and to bridge the gap to the diagnosis, treatment, and prevention of these diseases. We also suggested that the onset of hypertension, dyslipidemia, IgG4emia, and hyperuricemia has the characteristics of intestinal dysbiosis that induce them, and that correcting the increase/decrease in the abundance ratio of specific bacteria that are central to dysbiosis can lead to the suppression.

Academic Significance and Societal Importance of the Research Achievements

近年、生活習慣病をはじめとする全身疾患の発症リスクを高める原因として腸内細菌叢のディスバイオーシスの可能性が考えられつつあるが、腸内細菌叢のディスバイオーシスがこれらの疾患の発症リスクを高めるのかを解析した報告はない。本研究では機械学習や種々の統計解析手法を駆使して、腸内細菌叢のディスバイオーシスが高血圧、脂質異常症、IgG4血症、高尿酸血症の発症リスクを挙げること、そして各疾患発症リスクを挙げるディスバイオーシスの形態を特定できることを明らかにした。この結果より、これら4つの疾患の診断ならびに治療効果の評価に腸内細菌叢のディスバイオーシスを用いることが有用であることを示唆した。
