Outline of Research at the Start

エンハンサーは、遺伝子の発現スイッチとして働き、活性化したエンハンサーからは、enhancer RNA (eRNA)と呼ばれるノンコーディングRNAが合成されている。本課題では、eRNAの全長構造を読み解き、エンハンサーが機能するための塩基配列ロジックを発見し、エンハンサーの配列変異がゲノム制御やヒト疾病に及ぼす影響を解明する。

Outline of Annual Research Achievements

最終年度は、エンハンサー配列とヒト疾患に関連するDNA多型・変異との関係性を解析し、ヒト疾患の発症メカニズムの理解やエンハンサーの機能発現の解析を行った。このために、疾患発症に関連しているDNA多型・変異とエンハンサー配列を統合的に解析する。ゲノムワイド関連解析により同定されたDNA多型・変異の約90%はノンコーディング領域に存在しており、たんぱく質をコードしているエクソンに位置するのはごく一部である。エンハンサーに位置するDNA多型・変異は、eRNAの転写、転写因子結合などに影響を与え、遺伝子発現の調節異常につながることを示した(Oguchi et al. Science 2024)。機能的なDNA多型・変異を絞り込み、そしてそれらのエンハンサーや標的遺伝子への影響をマッピングし、ヒトの疾患発症メカニズムの理解に繋げた。また、ゲノム編集技術を用いて、機能的な検証実験を行った。期間全体を通して、申請者らが独自に開発してきた短鎖NET-CAGE法(Nature Genetics 2019)に申請者らの所属施設に設置されたPacBio長鎖シークエンシング技術と組み合わせる新しい実験手法を確立し、ライブラリー作成条件を最適化する実験を行った。これによりeRNAの転写開始点のみならず、スプライシング部位、転写終結点を決定することに成功している。開発された新技術を、ヒトの細胞株や組織に適用した。そして、さらにはエンハンサーとヒト疾患関連する変異・多型との統合的な解析を実施した。このようにBhagat博士らと共同して、本課題では、新規技術の確立によりeRNAの構造を読み解き、エンハンサーの配列変異がゲノム制御やヒト疾病に及ぼす影響を解明した。
