病害虫の認識は植物免疫機構において最も重要なステップである。本研究は植物免疫を誘導する病害虫由来物質の同定と、それを認識する植物の受容体を同定することを目的としている。植物の病害虫認識機構を分子レベルで理解することができれば、病害虫に対する植物免疫機構の全貌解明への大きな糸口となるだけでなく、新規の病害コントール法の確立につながる。

Outline of Annual Research Achievements |
I investigated the origin and evolution of pattern-triggered immunity (PTI) in plants. As mentioned, plants perceive PAMPs via PRRs to activate PTI. The origin and evolution of PRRs and PTI-signaling components is unclear. I performed a large-scale comparative genomic analysis on PRRs and PTI-signaling components in 350 plant species. While RLPs are generally more ancient than RLKs, RLK families have expanded much more than RLP families. I also showed that multiple PTI-signaling components are ancient and most show limited expansion in the plant lineage. These results suggest that PRRs and PTI signaling network are ancient and likely to exist before emergence of land plants, which then have evolved to become more complex and immune-specific in land plants.

1: Research has progressed more than it was originally planned.
Reason
The screening proposed last year is working well. More than 200 receptor candidates have successfully been cloned and they expressed well in plants. I have also tested and optimized the sensitivity of receptors to different pathogen extracts, such as bacteria and nematodes. The screening for PRRs that recognize pathogen extracts is still on-going, but there are already promising candidates of immune receptors against pathogens.
I have also finished a new project on the origin and evolution of pattern-triggered immunity. The manuscript for this work is under preparation and will be posted for publication within 2023.

Strategy for Future Research Activity |
The research project is on-going and progressing as planned. The screening is on-going and should be finished by the end of the year. The identified receptors will be further characterized and tested. I will continue to work on the identification of new receptors that confer resistance against pathogens. For the evolution project, the manuscript will be submitted by the end of 2023.
