Outline of Research at the Start

本研究では、人間活動によって汚染された土壌に生育する植物が土壌微生物と持つ関係性について理解すること、さらに、その土壌微生物の利用によって土壌の健全化および植物の生育促進が可能か明らかにすることを目的とする。具体的には、「汚染によって植物の生育や健康と関係の深い土壌微生物がどのような影響を受けるのか」、「汚染下でも健全に生育する植物の周辺土壌に生息する微生物群集の持つ特徴は何か」、「土壌移植や資材の投入などによって土壌微生物群集はどのように変化するのか、また、植物生育を助けるような微生物群集の形成を誘導することはできるのか」、などについて明らかにする。

Outline of Annual Research Achievements

研究開始時期に新型コロナウイルスによる影響でザンビアへの渡航が不可能だったなど複数の事情により、鉛・亜鉛汚染に限定せず、土地利用変化や環境変動に伴うストレス下における植物―土壌微生物間の相互作用の理解、および、植物生育の健全化に寄与する土壌微生物の機能についての研究を進めている。特に、ザンビアから輸入した鉛・亜鉛汚染の影響を受けた土壌、アメリカ・ボストンにおける都市化による土壌汚染の影響下にある土壌、アメリカ・ニューハンプシャーにある温暖化に伴い土壌凍結ストレスが増加した土壌、北海道の化学肥料・農薬を用いない水田と比較した慣行農法下にある水田土壌などを対象に研究を行っている。これまでの研究で、都市化、それに伴う林縁部の増加によって、植物と共生関係にある外生菌根菌の土壌に占める割合が低下することがわかったが、当該年度の研究により、外生菌根菌の活動の中でも特に窒素吸収の活動が林縁部で低下することが、土壌メタトランスクリプトーム解析により明らかとなった。この外生菌根菌の窒素吸収の活動と、樹木の葉の窒素濃度には相関があり、外生菌根菌が樹木の健全性において果たす役割の重要性を強調している。また、これまでの研究で、土壌の菌根菌の量と逆相関して土壌で植物病原菌および動物病原菌が増加することが明らかとなっていたが、当該年度において、生態系内の様々な微生物の生息地からサンプルを採取した結果を解析した結果、生態系内の異なる生息地間の間で病原菌の量がある程度維持されることが新たにわかった。これは、環境変動などによる土壌微生物叢の変化が生態系内の他の微生物叢にも影響を与えうる可能性を示唆している。このことから、ストレス環境下において、植物共生菌を維持し病原菌を減らす形で土壌微生物叢を健全化する手法を解明することで、生態系全体に生息する病原菌の低下にも繋がるかもしれない。

Reason

研究開始時期に新型コロナウイルスによる影響により渡航ができず、研究の方向性を大きく修正することになったが、その中でも、現地から輸入した土壌や、鉛・亜鉛濃度を変化させた土壌を用いて研究を進めてきた。また、鉛・亜鉛汚染に限定せず、臨機応変に様々なストレス環境下にある土壌を対象に切り替え、各ストレスがどのように土壌微生物、特に植物と共生関係を持つ微生物の生態や機能を変化させるのか、また、ストレス環境下において健全な環境由来の土壌微生物を導入することにより植物の生育や健康が改善するのか、についての研究を順調に進めている。土壌メタゲノム解析および土壌メタトランスクリプトーム解析技術を習得し、土壌微生物の機能のより多角的な理解に努めている。学会やセミナーでの講演および論文の発表も精力的に行っている。

Strategy for Future Research Activity

ザンビアから輸入した土壌の分析を進め、生育良好な個体周辺に共通する特異的な微生物要素についての分析を進める。また、室内実験により、鉛・亜鉛および有機物の添加に対する土壌微生物群集の応答に関するデータを集めており、これの解析を進める。特に、汚染下で低下する栄養塩循環に関る微生物の量や組成を特定し、植物の存在下で増加すると考えられる有機物が、これらの微生物要素を回復させるのか明らかにする予定である。加えて、重金属汚染度とも関連する都市―郊外傾度を用いた研究から、林縁部の環境において外生菌根菌が発揮する機能が変化することが土壌メタトランスクリプトーム解析により明らかとなってきた。外生菌根菌と入れ替わりで増加した病原菌の発揮する機能についても解析を行う。さらに、劣化土壌に複数タイプの森林の土壌を加えることで、土壌微生物群集の機能および植物の生育が改善されるのか観察するポット実験を開始したため、定期的に土壌を採取して経過を観察する予定である。また、それぞれ異なる生息域由来の真菌と細菌を混合し、群集の再構築過程を観察する実験を行っており、分析まで完了させた。この実験の解析についても進め、結果を統合して、外部から持ち込んだ微生物が、元々いた微生物とどのように群集を再構築するのか、さらにその新しい微生物群集は植物にとって有用な機能を持つのか、について解明する。これらの研究成果を引き続き随時論文としてまとめていく予定である。
