Outline of Research at the Start

肺は人間にとって必要不可欠な臓器であり、慢性閉塞性肺疾患(COPD)や特発性肺線維症など多くの呼吸器疾患は、有効な治療法がない致死的な疾患である。 本研究では、日本人多施設コホートや欧米諸国で蓄積された大規模データベースを用いてCOPDやIPF、COVID-19を対象としたゲノムワイド関連 解析結果を主軸として、末梢血や肺組織のトランスクリプトームを統合することで、病態の原因となる遺伝子(群)と組織間の相互作用を解明する。上記複数疾患を対象に包括的マルチオミクス解析を実践することで、複数疾患や人種横断的に共有された病態の根幹に関わる分子を同定し 、新たな治療薬の開発を目指す。

Outline of Annual Research Achievements

特発性肺線維症約100 名約200個の血液検体と、健常者約50個の血液検体からRNAを抽出し、全トランスクリプ トーム解析を実施した。IPFと健常者の間で発現変動遺伝子解析(DEG解析)を実施することで、IPF特異的に発現が亢進している遺伝子群を明らかとした。さらにエンリッチメント解析によって重要なパスウェイの同定を行なった。

また、既存の選択的RNAスプライシングとCOVID-19重症化/疾患感受性に対するゲノムワイド関連解析に対してメンデルランダム化解析、選択的RNAスプライシング とCOVID-19の因果関係を推定した。肺でのOAS1、ATP11A、DPP9、NPNTの選択的RNAスプライシングがCOVID-19重症化に関わっていることが推定された。また肺での MUC1とPMF1の選択的RNAスプライシングはCOVID-19疾患感受性に関わっていることが推定された。 本研究は筆頭著者としてNature Communicationsに発表した。

さらに、前年度より継続して、LongCOVIDに対する国際共同研究(ゲノムワイド関連解析)を実施する国際コンソーシアム(LongCOVID Host Genetics Initiative)を 主導しており、11コホートより集積した症例3,018名、コントロール994,582名を対象としたゲノムワイド関連解析のメタ解析を実施し、FOXP4遺伝子近傍の rs9367106:G>Cを有する場合にLongCOVIDの発症リスクが高いことがわかった(オッズ比 1.63, p=1.8x10-10)。本研究は現在論文推敲中である。

Strategy for Future Research Activity

特発性肺線維症と健常者の血液から得られた全ゲノムシークエンス結果と全トランスクリプトーム解析結果を統合し、以下の下流解析を進めていく。 1) 特発性肺線維症と健常者の遺伝子発現変動解析
2) 特発性肺線維症の進行例/非進行例間で比較した遺伝子発現変動解析
3) ゲノム情報と遺伝子発現量の関連解析
