Outline of Research at the Start

過去のヒトの居住環境を、古代土壌DNAを用いて分析・復元する手法を確立する。具体的には 、遺構の各区画(炉・トイレ・ゴミ捨て場・柱跡 ・水田跡など)から土壌を採取し、DNA抽出・配列解析(ショットガンシーケンス)を行う。これによって、各区画の使われ方によってDNA構成に どのような違いが出てくるのか(各区画のマーカーとなるDNAの探索)、古代土壌DNAから当時の食物・住居の素材・寄生虫などの衛生環境の情報 が得られるのか、を検証する。

Outline of Annual Research Achievements

過去のヒトの居住環境を、古代土壌DNAを用いて分析・復元する手法を確立する。具体的には、遺構の各区画(炉・トイレ・ゴミ捨て場・柱跡など)から土壌を採取し、DNA抽出・配列解析(ショットガンシーケンス)を行う(図5)。これによって、(1)各区画の使われ方によって、DNA構成にどのような違いが出てくるのか(各区画のマーカーとなるDNAの探索)、(2)古代土壌DNAから、当時の食物・住居の素材・寄生虫などの衛生環境の情報が得られるのか、を検証する。この手法を複数の遺跡に適用し、その有効性を検証するとともに、最終的には、古代土壌DNA解析から当時の居住環境を復元することを目指す。
最終年度及び研究期間全体においては、目的とすることをある程度の水準で達成することができた。(1)に関しては、特に家畜のDNAが土壌から検出された遺跡において、その家畜の腸内細菌を検出することができ、家畜をその場所で飼育していたことが明らかとなった。また、(2)に関しては、キャンプサイトの遺跡から寄生虫のDNAが検出されたほか、住居跡からはその建築材として用いられた可能性の高いものが発見された。
詳しい報告については、各遺跡の報告書及び学術論文として公表していく予定である。また、将来的には分析する遺跡数を増やすことにより、この結果をより信頼性の高いものとすると共に、花粉分析のように土壌DNAも考古学の手法の一つとなることを目指していたが、本研究においてはその礎が形成されたといえる。
