Outline of Research at the Start

肥満細胞（Mast cell;MC）は組織常在性の免疫細胞で、マウスでは結合組織型MC（CTMC）と粘膜型MC（MMC）の2つのサブセットに分類され、皮膚MCはCTMCである。CTMCは定常状態ｆｒは自己増殖により局所に維持されることが明らかになっている。しかし、アトピー性皮膚炎（AD）のような炎症状態において、皮膚MCのようなCTMCがどのように維持されるかは、まだ十分に解明されていない。我々は、AD様皮膚において、皮膚MCが、常在MC（rMCs）の局所増殖と骨髄由来肥満細胞前駆細胞の浸潤・増殖の双方によって維持されていることを明らかにした。

Outline of Annual Research Achievements

1．マウス骨髄由来誘導性肥満細胞におけるCD9の機能解析
RNAseq及びフローサイトメトリーを用いた実験結果より常在型肥満細胞と比較して、骨髄由来誘導性肥満細胞がCD9を特異的に発現していた。さらにはCD9のリガンドであるIL-16 は正常皮膚に比較してアトピー性皮膚炎で上昇していた。そこで誘導性肥満細胞はCD9-IL-16依存性にアトピー性皮膚炎病変部へ浸潤しているのではないかと仮説を立てた。しかしながら、IL-16 阻害抗体及びCD9KOマウスを用いても、誘導性肥満細胞の浸潤には変化を認めなかった。さらには、CD9KOマウスの骨髄より肥満細胞を誘導し（BMMC）、細胞増殖速度、細胞表面マーカーの発現を確認したが、WTマウスのBMMCと同程度であった。以上より、骨髄由来誘導性肥満細胞はCD9を特異的に発現しているが、皮膚浸潤及び肥満細胞の文化においては重要でないと考えられた。
2．制御性T細胞の関与
これまでの既報よりCD25陽性CD4T細胞が肥満細胞前駆細胞の末梢臓器への浸潤へ関与していることが示唆されていた。制御性T細胞を後天的に除去可能なFoxp3DTRマウスを用いて、アトピー性皮膚炎を誘導した。Foxp３DTRにDT投与したマウスでは耳介の炎症は増悪を認め、骨髄由来誘導性肥満細胞の浸潤には変化を認めなかった。他の抹消臓器とは異なり、皮膚では制御性T細胞は肥満細胞の浸潤に重要でないと考えられた。
