Outline of Research at the Start

昆虫のオス殺し細菌Wolbachiaの進化・オス殺し分子メカニズムに及ぼすファージWOの影響を解明する。農業・衛生害虫の防除への応用が期待されているWolbachiaは、オスのみを致死させるオス殺しを引き起こす。しかしWolbachiaがオスを殺す機構:原因遺伝子、分子機構、およびオス殺しを獲得した進化経緯は明らかでない。本研究では、チャ樹害虫チャハマキから同定されたオス殺しWolbachiaを操作するファージWOをもとに、オス殺し原因WO遺伝子を同定し、昆虫の遺伝子発現解析から分子メカニズムを解明し、野外調査およびゲノム・進化解析からWolbachiaの進化にWOが及ぼす影響を解明する。

Outline of Annual Research Achievements

昆虫に感染する共生細菌Wolbachiaが引き起こすオス殺し現象の分子メカニズムおよび獲得プロセスを、Wolbachiaに感染するファージWOに着目して解明することを本研究の目的とした。2023年度は、オス殺しWolbachiaがもつ遺伝子の機能解析に加えWolbachiaのゲノムを解析した。一連の研究活動を通し、Wolbachiaは進化の過程で複数のオス殺しメカニズムを獲得したこと、ファージWOがWolbachiaのオス殺しの進化に保深く関与してきたことが浮き彫りになった。具体的な成果は以下のとおりである。

・複数のオス殺し遺伝子:ショウジョウバエおよびチャハマキを用いてWolbachia遺伝子の機能を解析した。チャハマキオス殺しWolbachia wHm-t株が持つオス殺し関連ファージ領域座上遺伝子:wmkおよびHm-Oscarを強制発現させたところ、wmk遺伝子はショウジョウバエのオスを殺した一方でチャハマキには作用せず、逆にHm-Oscarはチャハマキのオスを殺した一方でショウジョウバエのオスを殺さなかった。これらの結果から、Wolbachiaが複数の昆虫オス殺し遺伝子を持つことが示唆された。

・Wolbachia比較ゲノム解析:様々な昆虫由来のWolbachia十数株のゲノムを解読、環状ゲノムを構築した。多くのチョウ目オス殺し・メス化 Wolbachia 株間では、メイガ類およびチャハマキのオスを殺すOscarホモログが保存されていた一方、ハエ目昆虫および一部のチョウ目由来のオス殺し Wolbachia からはOscarホモログが検出されなかった。また一部のオス殺し・メス化 Wolbachia から、wHm-t株のオス殺し関連ファージ領域が検出された。以上から、オス殺しやメス化に関わる複数の遺伝子の存在とWolbachiaの進化へのファージの関与が示唆された。
