Outline of Research at the Start

本研究の目的は、ネッタイシマカのデングウイルス感染・複製への抵抗性を決めている遺伝子をマーカーとして、デングウイルス伝播ポテンシャルが高い蚊地域集団を見つけ、それらを狙い撃ちにした殺虫剤散布などの効率的なデング熱制御手法を提案することである。東南アジアのデング熱流行都市で野外調査を行い、ネッタイシマカ地域集団のウイルス抵抗性遺伝子の有無とデングウイルス感染率・体内濃度の関係などを検証することで、抵抗性遺伝子に着目した効率的なデング熱制御手法を提案する。

Outline of Annual Research Achievements

今年度は、以下二つの研究項目を中心に研究を進めた。

研究項目2 蚊地域集団のウイルス抵抗性遺伝子の多様性評価(フィリピン・インドネシアとの共同研究):メトロマニラとバンドン近郊のチマヒ市、各世帯の屋内に誘虫紫外線ランプと吸引ファンを備えた電動蚊採捕機を設置し、ネッタイシマカ等の多様な蚊成虫を採取した。採取した蚊サンプルは形態学的に同定した後、RNAlaterに個体ごとに入れて冷凍庫内に保存した。メトロマニラで採取・保存したネッタイシマカ個体から十分な量のDNAを抽出するためのプロトコルを確立し,ウイルス抵抗性遺伝子を含む様々な遺伝子をゲノム全体からランダムに選択して、それらの遺伝的多様性を調べた。具体的には、次世代シーケンサーを使ったddRAD解析により、約1000遺伝子座を検出し、メトロマニラ内の地域個体群の遺伝的多様性ならびに地域個体群間の遺伝的差異を調べた。

研究項目3「蚊地域集団のデングウイルス感染率・体内濃度と抵抗性遺伝子の関係の検証(フィリピン・インドネシアとの共同研究)」:研究項目2で採取・保存した蚊個体の一部の個体を使って、個体レベルから十分な量のRNAを抽出するためのプロトコルを確立した。インドネシアのバンドンに設置した愛媛大学ーパジャジャラン大学国際共同研究ラボラトリーにおいてデングウイルスの1~4型をそれぞれ定量PCRで検出する実験手法を最適化し、現地でデングウイルスを検出・定量できる体制を整備した。

Reason

研究項目2(蚊地域集団のウイルス抵抗性遺伝子の多様性評価)、研究項目3(蚊地域集団のデングウイルス感染率・体内濃度と抵抗性遺伝子の関係の検証)共にインドネシア、フィリピンにおける蚊採取は順調に行われ、また、採取した蚊から十分な量の核酸(DNA,RNA)を抽出して、デングウイルスを現地で定量・検出するプロトコルを確立することができた。以上より、概ね順調に進展しているを評価した。

Strategy for Future Research Activity

研究項目1(蚊ゲノムのウイルス抵抗性遺伝子の網羅的探索):標的遺伝子を除去する酵素Cas9を発現するネッタイシマカ培養細胞系を構築する。
研究項目3(蚊地域集団のデングウイルス感染率・体内濃度と抵抗性遺伝子の関係の検証):フィリピンのマニラ首都圏とインドネシアのバンドンを対象にして、ネッタイシマカ体内のデングウイルス感染率・体内濃度と抵抗性遺伝子の関係を検証する。
