Outline of Research at the Start

酵素がもつ高い分子識別能と,遷移金属錯体の汎用的で多様な機能を相乗的に活用し,単独の触媒では成しえない難易度の高い変換を1工程で,または1つの容器内で進行させる。本法は,高い選択性,効率性と拡張性を備えた環境低負荷の革新的な物質生産・変換法となる。本研究によって,第三級アルコール,多環式複素環,軸不斉ビアリール化合物,メビウス様分子,重水素化物質などの多様性に富む高機能性有機化合物の短段階合成を可能にする。

Outline of Annual Research Achievements

入手容易なラセミ体やアキラルな化合物を原料として、光学的に純粋な分子や高機能性分子に100%変換する方法、環境負荷の少ない分子合成法や分子変換法を創出することを目的として、日独間で国際共同研究を行った。本年度は以下の成果を得た。
1.第3級アルコールのリパーゼ触媒による動的速度論的光学分割(DKR)の適用拡張と反応時間短縮を図った。日本側は塩基の添加がリパーゼ触媒光学分割の反応速度を向上させることを見出し、適用基質を拡張させた。ドイツ側は触媒活性が向上したリパーゼ改変体の作成に成功した。また、日独共同でラセミ化触媒を必要としない軸不斉ビアリール化合物のリパーゼ触媒DKR法を開発した。さらに、この概念の適用拡張を図るべく、大阪大学の院生がビーレフェルド大学に滞在し、リパーゼ改変体の作成研究を行っている。また、赤井はビーレフェルト大に滞在し、同大学の研究メンバーとリパーゼ触媒DKR法の今後の研究展開に関して打合せを行った。
2.重水素を有するラセミ体第2級アルコールにリパーゼ触媒DKR法を適用し、光学的に純粋な重水素化アルコールに高収率・高重水素化率で変換できることを実証した。
3.ブレンステッド酸触媒存在下、水を求核剤とするヘテロ環形成反応を開発し、環状アセタールやオキサシラサイクル構造を有するスピロシクロブタン誘導体の合成に成功した。
4.キラルバナジウム触媒を分子間および分子内酸化カップリングに適用することで、メビウス型アヌレンを単一エナンチオマーとして合成した。また、笹井はビーレフェルト大に滞在し、本研究に関して打合せを行った。
5. 電解酸化と酵素分割のハイブリッドプロセスによるデータ駆動型ダイバージェント不斉合成研究の基礎研究成果を論文発表した。

Strategy for Future Research Activity

1.リパーゼとラセミ化剤を併用する第3級アルコール類の動的速度論的光学分割において、両触媒の長時間共存性を更に改善するために反応場の分離法などを検討し、基質適用性と実用性に優れたDKR法を完成させる。
2.軸不斉ビアリール化合物のDKRのためのリパーゼ改変体を作成し、基質適用性の拡張を目指す。
3.窒素求核剤としてアニリンを用いるヘテロ環形成反応の開発に取り組み、任意の位置に窒素原子を有する多様な含窒素ヘテロ環の合成研究を推進する。
4.電解酸化と酵素分割のハイブリッドプロセスによるデータ駆動型ダイバージェント不斉合成研究をデータ駆動型不斉合成へと展開する。
