Outline of Research at the Start

酵素がもつ高い分子識別能と,遷移金属錯体の汎用的で多様な機能を相乗的に活用し,単独の触媒では成しえない難易度の高い変換を1工程で,または1つの容器内で進行させる。本法は,高い選択性,効率性と拡張性を備えた環境低負荷の革新的な物質生産・変換法となる。本研究によって,第三級アルコール,多環式複素環,軸不斉ビアリール化合物,メビウス様分子,重水素化物質などの多様性に富む高機能性有機化合物の短段階合成を可能にする。

Outline of Annual Research Achievements

入手容易なラセミ体やアキラルな化合物を原料として光学的に純粋な分子や高機能性分子に100%変換する方法、並びに、環境負荷の少ない分子合成法や分子変換法を創出することを目的として、日独の協働研究を行った。本年度は以下の成果を得た。
(1) a) メソポーラスシリカ、ピッカリングエマルション、ポリジメチルシロキサンチューブなどによる反応場分離技術を活用し、リパーゼ触媒による第3級アルコールの動的速度論的光学分割(DKR)法を改良し、適用を拡張した。また、ドイツ側研究者によって作成されたリパーゼ改変体を用いることで、より嵩高いアルコールにも適用できることがわかった。b) 日独共同でラセミ化触媒を必要としない軸不斉ビアリール化合物のリパーゼ触媒DKR法を開発した。c) 上記a, bのテーマを実施するため、大阪大学の大学院生がビーレフェルド大学に滞在し、リパーゼ改変体の作成研究等を行った。
(2) LED光照射下、2-ナフチルアミンと2-ナフトールとの酸化的カップリング反応にキラルバナジウム触媒を用いると2-アミノ-2’-ヒドロキシ-1,1’-ビナフチル誘導体が最高95%収率・80% eeで得られることを見出した。本反応ではホモカップリングはほとんど進行せず、ヘテロカップリング体が効率的に得られる。
(3) 有機分子光触媒として機能する非対称ヘテロ[8]サーキュレンのワンポット電解合成法を確立した。本サーキュレン光触媒にLED光を照射すると様々なクロスカップリング反応がメタルフリーにて進行する。機械学習を活用することで、光触媒能及び光学特性などの機能特性を予測することにも成功した。
(4) アニリンを求核剤とするビシクロブタン誘導体の環形成反応の開発に成功し、ジヒドロイソキノリン構造を持つ新規スピロシクロブタン類の合成を達成した。

2: Research has progressed on the whole more than it was originally planned.

Reason

(1) 第3級アルコールのリパーゼ触媒DKR法の適用拡張研究と、軸不斉ビアリール化合物のDKR法の開発研究が当初の計画通り進行した。これらの成果をまとめ、論文を4報発表し(改訂中を含む)、2報作成中である。
(2) 2-ナフチルアミンと2-ナフトールとのラジカルカチオン-ラジカルカップリング反応を確立することでホモカップリング副反応を防ぐごとに初めて成功した。現在論文投稿準備中。
(3) 非対称サーキュレンのワンポット電解合成のモデル研究であるヘテロヘリセンのワンポット電解合成(Advanced Synthesis & Catalysis誌)の論文が、2023年に同誌に掲載された最も閲覧された論文の上位10%(Top Viewed Article in Wiley)にランクインした。
(4) 多様なヘテロ環を有するスピロシクロブタン類の合成において、含ケイ素化合物や含窒素化合物などの合成に成功し、当初の計画通りに進行した。

Strategy for Future Research Activity

(1) 反応場分離法の更なる改良を行い、第3級アルコールのリパーゼ触媒DKR法の適用拡張を更に推進する。また、現在、新しい概念に基づく軸不斉ビアリール化合物のDKR法を開発中であり、これを完成させる。
(2) 光エネルギーとバナジウム触媒との協働的活性化研究を推進し、これまでラセミ反応ですら進行しなかった様々な不斉ヘテロカップリング反応を開拓する。
(3) ヘテロサーキュレン光触媒によるメタルフリーなカップリング反応のデータをデジタル化し、触媒能評価予測に機械学習を活用することで効率的に分子を再デザインする。可視光でも機能する光有機分子触媒の創製を目指す。
(4) 多様なヘテロ環を有するスピロシクロブタン類の合成において、これまでの成果は5・6員環ヘテロ環に限られていたが、今後は7・8員環の中員環ヘテロ環を有するスピロ化合物の合成に展開する。
