Outline of Research at the Start

運動時には骨格筋の酸素需要に応えるべく血圧は上昇する.その神経性調節機構の一つに活動筋からの反射(運動昇圧反射)が重要な役割を担う.特に,骨格筋の機械刺激により血圧が増大することを筋機械受容器反射と呼ぶ.それは,機械受容チャネルの活性化から始まるが,そのチャネルの特定は進んでいない.また,様々な病態による筋機械受容器反射の変容機序も不明である.本研究は,University of Texas Southwestern Medical CenterのDr. Mizunoラボと共同で,機械受容チャネルの同定を端緒に,総合生理学的アプローチにより運動昇圧反射のメカノバイオロジー機構全容解明を目指す.

Outline of Annual Research Achievements

筋機械受容器反射を担うチャネル分子の特定を第一の目的としている.候補チャネルが筋感覚神経にてメカノトランスダクションを示し,さらに運動昇圧反射に寄与することを明らかにする.
Transient receptor potential vanilloid 2 (TRPV2)が筋機械受容器反射に関与しているか否かの検討を行った.TRPV2の神経特異的ノックアウトマウスと,健常マウスにおいて除脳・無麻酔下において,下腿三頭筋のストレッチに対する血圧応答を比較した.しかし,除脳時の出血により,生理学的範囲の循環状態で測定できた例が少なく,継続して検討する必要がある.
本課題は,筋機械受容器反射における感作機序を明らかにすることを第二の目的としている.そのために感作を引き起こし得る骨格筋慢性アシドーシスを示すモデル動物を用いて検討した.温度差ストレスをラットに暴露することにより,暴露から少なくとも3週間は骨格筋組織pHが低下していることを確認した.骨格筋が酸性化していることから,酸に感受性を持つイオンチャネルであるAcid sensing ion channel (ASIC)3の関与を疑った.RCSラットでは,下肢動脈への酸性溶液に対する昇圧応答は対照動物に比べ有意に増強した.さらに,下腿三頭筋のストレッチに対する血圧応答はRCSにより増強したが,ASIC3の拮抗薬投与によりその増強は抑制された.以上より少なくともこの筋機械受容器反射における感作機序にASIC3が関与していることが機能面から示唆された.

Reason

2023年度にTRPV2の検討を終えることができなかったが,温度差ストレスによる骨格筋慢性アシドーシスを示す動物の筋機械受容器反射増強の機序の一部解明は達成できたため.

Strategy for Future Research Activity

目的2の機械感作の機構の一部を明らかにする.これまでに,慢性心・腎不全,末梢血管疾患,糖尿病などにより筋機械受容器反射が亢進するとの報告がある.これら病態ではアシドーシスが共通の症状としてある.酸に感受性をもつチャネルには,ASIC3の他にTRPV1がある.通常,TRPV1はメカノトランスダクションには直接関与しないと考えられている.そこで,このチャネルが機械閾値を下げる結果,メカノトランスダクションを獲得,あるいは関与するように変容することで,筋機械受容器反射が過剰亢進するという仮説を検証する.
骨格筋の慢性的なアシドーシスを呈するモデル動物(Sprague-Dawleyラット使用)を利用する.この慢性アシドーシスラットにおいて, ex vivo, in vivoのマルチスケールにおいて,TRPV1の作動薬の応答が増加することを電気生理学・循環動態学的に確認する.さらに,阻害剤にて骨格筋ストレッチによるメカニカルストレスに対する応答が減弱することを電気生理学・循環動態学的に検討する.
さらに,2023年度に終えることのできなかったTRPV2の検討も継続して行う.
