| Outline of Research at the Start |
全ゲノムシーケンサーを用いた1細胞遺伝子発現解析技術が発達し、初期胚やがん細胞、農作物のクローン個体など、遺伝的には均質な細胞集団であっても、個々の細胞の遺伝子発現がバラつき、不均質であることが次々と報告されている。しかし現在の研究は、生体組織や臨床検体で不均質性を網羅的に報告する「現象論」に留まり、不均質性が生まれる「分子メカニズム」の解明は進んでいない。申請者は、分裂酵母を用いた研究から、ゲノム全域の遺伝子発現不均質性促進・抑制因子を見出した。本計画では、これまでの知見をヒト細胞に応用し、腫瘍悪性度やHIVウイルス感染細胞のウイルス増殖能の不均質性の制御関連因子をスクリーニングする。
