Research Abstract |
NF-kappaB signaling and interferon production pathway play a pivotal role on biological defense system in various spices. In this study, we identified that SHARPIN is an additional subunit of LUBAC, which produces a novel type of linear polyubiquitin chain and regulates NF-kappaB pathway. Genetic deficiency of SHARPIN causes severe inflammation such as dermatitis in mice. Furthermore, we showed that A20, a deubiquitinatinase, involves evolutionary conserved linear ubiquitin specific binding site at 7th ZF domain. Moreover, we analyzed structure-function study of virus DNA sensor protein, cyclic GMP-AMP synthease (cGAS).
