Research Abstract

血友病Ａ遺伝子治療：In vivoへのベクターの直接投与には染色体への組み込みがほとんどおこらないアデノ随伴ウイルス(AAV)ベクターを用い、ex vivoにて細胞への遺伝子導入し再移植する遺伝子細胞治療には染色体への組み込みが必要であるためSIVベクターを用いることとした。ヒトに近い種属の血友病モデル動物（血友病Ａクローンブタ）の作製に成功した。血友病Ａブタは出生後から出血傾向を示し、遺伝子治療効果判定に優れたモデル動物である。遺伝子細胞治療として、SIVベクターをもちいて間葉系幹細胞に導入し、血友病マウスへ移植し治療効果を確認出来た。血友病Ｂ遺伝子治療：マウスでの検討を終え、カニクイザルを用いた前臨床実験を遂行している。AAV8ベクターの遺伝子導入を阻害する中和抗体陽性サルでマイクロカテーテルを用いた門脈内ベクター投与を行い３頭のサルにおいて治療域に達する導入遺伝子由来FIX発現が得られ、抗AAV8抗体が存在していても遺伝子治療が可能であることが示された。血友病Ｂ遺伝子治療臨床研究に向けて作製したGMPグレードヒトFIX遺伝子搭載AAV8ベクター(GMP-AAV8hFIX）の純度および生物学的活性を検討したところ、SDS-PAGEにてGMP-AAV8hFIXの純度は極めて高く、電子顕微鏡による解析でもempty vectorの混入は低く抑えられていた。生物学的活性は血友病Ｂマウスを用いた検討では5x10の11乗/kgで平均58％の活性発現がえられマウス血漿中のFIX蛋白濃度とFIX活性は一致し、GMPグレードベクターの遺伝子導入活性が確認できた。
