Research Abstract

扁形動物プラナリアは,多能性幹細胞ネオブラストが全身の30%の細胞を占めるために高い再生能をもつが,その生殖様式には①生殖器官を持たず,分裂と再生のみで無性的に殖えるもの, ②生殖器官をつくり2匹が対合して有性的に殖えるもの, そして③その両者を環境により転換させるものの3種類が共存する.また,核型には,2倍体, 3倍体といった倍数性とともに異数性, 混数性など染色体の変化が数多く観察されている.
本課題に用いたリュウキュウナミウズムシDugesia ryukyuensisでも核相は2n = 14の2倍体と3n = 21の3倍体が混在し,さらに上記の3種類の生殖様式が共存する.よって,この種では生殖器官をもつ3倍体の個体も存在するが,雌性生殖細胞のみの単為発生によりゲノムを混合することなく子孫を作っていると考えられてきた.申請者は生殖器官をもつ個体の卵巣域,精巣域の染色体観察から,相同染色体が対合し,相同染色体間で乗り換えを起こしたキアズマ構造を確認した.
本課題で,申請者は個体ごとのゲノムを識別できるマイクロサテライト遺伝子をマーカーとして用いて親子鑑定を行い,分子レベルでゲノムが混合されていることを示した.これらのことは,減数分裂によりできた卵と精子が受精し,ゲノムが混ぜ合わされて次世代を作っていることを示した.また,2匹の3倍体有性生殖個体が接合して産んだ卵殻から,2倍体の仔虫と3倍体の仔虫が同時に誕生するが,これらの減数分裂時の染色体の詳細な観察から,雄性生殖細胞では第1減数分裂前期で既に2倍体になっているが,雌性生殖細胞の第1減数分裂前期では2価の染色体と1価染色体が共存しており3倍体の状態である事が示された.このようにプラナリアでは雌性と雄性における減数分裂の機構が異なることが示唆された.
