ヒト臍帯血造血幹細胞の真に安全な新規増幅法開発

Research Abstract

【背景・目的】これまでマウス移植系でHoxB4遺伝子導入による造血幹細胞(HSC)の増幅効果が報告されてきた。一方、HoxB4発現レトロウイルスベクターを用いたサル・イヌの大型動物移植系では、マウスでは観察されなかった白血病の高率な発症が示され、安全性が危惧されている。HoxB4は短期間発現で増幅効果を呈するのに対し、長期間発現で白血病を惹起するとされ、そこで我々はセンダイウイルス(SeV)ベクターのうち特にポリメラーゼ遺伝子欠損型SeV(SeV/ΔP)ベクターに注目した。これはウイルスの自己複製能が欠損しておりex vivoでの一過性発現を可能にする。今回このSev/ΔPベクターを用いたHoxB4一過性発現によるヒトHSC増幅に関する有効性と安全性をヒツジ移植系で検証した。
【方法】ヒト臍帯血よりCD34^+分画を分離後、HoxB4遺伝子発現SeVベクターを96時間で計4回感染させた。Sev/ΔP/HoxB4感染細胞は、免疫系が未成熟な45-50日齢(満期147日)のヒツジ胎仔腹腔内へ移植した(HoxB4群;n=4,対照群;n=4)。生後の仔ヒツジ骨髄サンプルを用いたコロニーPCRによりヒト由来造血細胞の割合(ヒトキメラ率)を定量評価した。また定期的な末梢血・骨髄評価により白血病発症の有無をモニターした。
【結果】SeVベクターによるヒトCD34^+細胞への遺伝子導入効率は約70%(MOI=100)で効率良く導入可能であった。10^5個のCD34^+細胞を移植した場合、生後の仔ヒツジ骨髄中のヒトキメラ率は、HoxB4群では対照群の4.8倍と計算された。なお移植後40ヶ月時点で白血病発症は皆無であり、仔ヒツジ骨髄・末梢血中に残存SeVベクターゲノムはPCRレベルで検出されなかった。
【結語】SeV/ΔPベクターにより、ヒトCD34^+細胞への高い遺伝子導入効率を保ちつつ一時的遺伝子発現が可能となった。SeV/ΔPベクターによるHoxB4の一過性発現の結果、ヒツジ胎仔移植の評価系ではヒトHSCが有効に増幅されると同時に安全性も担保されていると考えられた。
