Research Abstract |
We transferred full-thickness human skin grafts onto SCID mice and simultaneously injected the PPP patients’ tonsillar mononuclear cell (TMC)or peripheral blood mononuclear cells (PBL) intraperitoneally. Four weeks after the transfer, we analyzed the localization of the lymphocytes in the skin graft with immunohistochemical method. We also investigated the value of human IL-6, IL-17, human IFN-γ、human TNF-α、the human anti-HSP60 antibody of lymphocytes in the murine sera. In the 63.6% of the SCID mice transplanted with the TMC of patients with PPP, CD4+ T lymphocytes infiltrated the area around the papillary dermis of their skin grafts. But in the group of the mice injected with the PBL of patients with PPP, only a few or no infiltrating CD4+ T lymphocytes were showed around the papillary dermis of their grafts . We detected anti-HSP60 antibody and human IL-17, IL-6 more frequently and with higher levels in the serum of PPP TMC/PPP skin SCID mice and PPP TMC/SCID mice with HSP60 treatment than other controls.
