Research Abstract

プロゲステロン(P4)は妊娠の成立・維持に必須であり、P4受容体(PR)を介して作用する。子宮収縮抑制や免疫抑制などの多様な作用が知られているがその機序の詳細は未だ不明である。PRコシャペロンFKBP52欠損マウスは子宮内膜のP4応答能低下するもののP4投与により着床以後の妊娠過程を観察でき、P4に対する応答能が低下して十分なP4の作用を得るには大量のP4の付加が必要な状態、いわゆる、「P4抵抗性」の有用なin vivoモデルと考えられる。本研究ではこのマウスを用いて、妊娠に最も重要な因子とされるP4の妊娠成立・維持にどのような経路や物質を介して働いているかという点を明らかにすることを目的とした。卵巣摘出・P4投与した野生型マウス、FKBP52欠損マウス、PR欠損マウスから子宮を採取し、プロテオミクス解析を用いてFKBP52欠損子宮およびPR欠損子宮で低下しているgalectin-1(Gal1)という糖鎖結合蛋白を同定した(Hirota Y,et al.Endocrinology 2012)。In vivoの検討により、Gal1は妊娠子宮においてP4依存的に誘導される分子と考えられた。野生型マウスの妊娠子宮において、Gal1は着床部位の子宮内膜問質細胞(脱落膜細胞)に強く発現していた。ヒト子宮内膜間質細胞初代培養系でも、P4を用いたin vitroの脱落膜化によってGal1発現が増加した。またFKBP52欠損マウスではP4の投与量が少ない場合に妊娠中期の着床部位への白血球遊走および流産が増加したが、Gal1の補充によってこの表現型が救済できた。Gal1についての近年の報告では、T細胞のアポトーシス促進、IFNγやIL2などのThlサイトカイン分泌抑制、免疫寛容への関与が示されており、妊娠子宮においてもGal1がP4の炎症抑制作用や妊娠の維持機構の一端を担っている可能性が考えられた。
