Outline of Research at the Start

タンパク質は人工の分子では成し得ない高度な機能を有する。したがって、タンパク質を理解することは生命現象の理解のみならず、高度な機能性分子の創成に重要な知見を与える。タンパク質の機能発現機構を理解するうえで重要な点は、構造変化によって複数の機能部位が互いに連動していることにある。この連動的構造変化を可能にする鍵として、タンパク質構造の高い空間充填が挙げられる。本研究では、タンパク質構造の稠密性を、アミノ酸置換および非天然補欠分子族の再構成によって操作する。これらの操作が、タンパク質の連動的構造変化および機能活性がどのように影響するかを定量的に調べ、稠密性の意義を明らかにする。
